急性百草枯中毒大鼠血清lL-10、TNF-α水平变化及黄芪注射液的干预作用

             

摘要

Objective To observe the dynamic changes of serum IL-10 and TNF-αin acute paraquat poisoning rats and the intervention effect of astragalus injection and to explore the therapeutic effect and mechanism of Astragalus injection in the treatment of acute paraquat poisoning.Methods 90 SD rats were randomly divided into three groups: control group,model group and treatment group,30 rats in each group. Each group was divided into 6h,12h,24h,72h,7d five time points,each time point for 6 rats. The model group and the treatment group given a one-time paraquat 25mg·kg-1 intraperitoneal injection exposure while control group with saline intraperitoneal injection. After exposure to 30 min,the treatment group was given intraperitoneal injection of Astragalus Injection(4 mg·kg-1),one times/day,and normal saline intraperitoneal injection with the same amount of control group and model group. The dynamic changes of serum TNF-αand IL-10 in each group at the corresponding time points were monitored by enzyme-linke immunosorbent assay(ELISA)in rats.Results The serum TNF-αlevel of model group at each time point were significantly increased compared with the control group(P<0.05)while TNF-αof the treatment group at each time point were significantly lower than that of model group(P<0.05). The level of IL-10 model group after injection in 12h began to increase,reached the peak at 72h,compared with the control group,the difference was statistically significant(P<0.01). The expression of IL-10 in the treatment group at the same time point were significantly lower than the model group(P<0.01). Conclusion The serum levels of IL-10 and TNF-αin rats with acute paraquat poisoning were increased and the protective effect of Astragalus injection on the lung tissue of acute paraquat poisoning was probably related to the decrease of serum IL-10 and TNF-α.%目的 观察急性百草枯中毒大鼠血清白介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)的动态变化以及黄芪注射液的干预作用,从而探讨黄芪注射液对血清IL-10、TNF-α的影响及作用机制.方法 90只SD大鼠随机分为三组,即对照组、模型组及干预组,每组各30只,每组又分为6h、12h、24h、72h、7d五个时间点,每个时间点取6只大鼠.模型组和干预组一次性给予百草枯25mg·kg-1腹腔内注射染毒,对照组用等量等渗盐水腹腔内注射,染毒后30 min,干预组给予黄芪注射液腹腔注射(4mg·kg-1),1次/d,对照组和模型组同时用等量等渗盐水腹腔注射.各组于相应时间点应用酶联免疫吸附法(ELISA)监测大鼠血清中IL-10、TNF-α的动态变化.结果 与同时间点对照组比较,模型组IL-10水平在染毒后12h升高(P<0.05),72h达到高峰(P<0.01),7d基本降至正常;黄芪干预组IL-10水平升高趋势与模型组相同,但在72h时干预组IL-10水平低于模型组(P<0.01),7d降至正常;大鼠染毒后,模型组和干预组TNF-α 水平各时间点均高于对照组(P均<0.01),72h达到高峰(P<0.01);干预组各时间点均较模型组降低(P均<0.05或<0.01),7d时仍高于对照组.结论 急性百草枯中毒大鼠血清IL-10、TNF-α上升,黄芪注射液可以降低急性百草枯中毒大鼠血清IL-10、TNF-α水平.
