目的 探讨复方芪珠片对大鼠四氯化碳(CCl4)慢性肝损伤及肝纤维化模型的保护作用.方法 纯种SD大鼠40只,随机分成正常组、模型组、中药大剂量组、小剂量组、秋水仙碱组.除正常组外,其余各组大鼠皮下注射40%CCl4橄榄油造模.中药大剂量组以16.2 g/kg剂量灌胃,中药小剂量组给药剂量为9.72 g/kg,秋水仙碱组用5 mL/kg剂量灌胃,模型组用同等体积的生理盐水灌胃,每日1次,于造模开始次日给药,连续7周.观测大鼠肝功能(ALT、AST、GGT)、肝纤维化指标(HA、PC-Ⅲ、LN)、肝脏组织学检查.结果 大剂量组、小剂量组、秋水仙碱组皆可降低大鼠血清ALT、AST数值,尤以中药大剂量组显著(P＜0.01).大剂量组和秋水仙碱组能显著降低HA、LN、PC-Ⅲ(P＜0.05).大剂量组显著改善肝脏脂肪变性和炎症坏死,减轻汇管区结缔组织增生和炎细胞浸润.结论 复方芪珠片对大鼠CCl4慢性肝损伤及肝纤维化模型具有保护作用,主要机制可能是通过减轻肝脏炎症反应.%OBJECTIVE To investigate the protective effects of Compound Qizhu Tablet on treating model rats with chronic liver injury and hepatic fibrosis caused by CCl4. METHODS 40 SD rats were randomly divided into normal group, model group, Qizhu high-dose group, Qizhu low dose group and colchicine group. All the rats except normal group received 40％ CCl4 subcutaneous injection. Rats in bith Qizhu high-dose group and small dose group were given gavage of 16.2 g/kg and 9.72 g/kg Qizhu decoction respectively, while rats in colchicine group were fed with 5 mL/kg colchicine.Rats in model group were offered the same volume of saline by gavage once a day for 7 weeks. Liver function (ALT, AST,GGT), hepatic fibrosis (HA, PC- Ⅲ, LN), and liver histology were observed. RESULTS ALT and AST in serum of Qizhu high-dose group, low dose group as well as colchicine group were reduced, especially those in high dose group. HA, LN and PC-Ⅲ significantly decreased in both high-dose group and the colchicine group (P＜0.05). The improvement of hepatic steatosis, inflammation and necrosis showed in Qizhu high-dose group which also reduced connective tissue proliferation and inflammatory cell infiltration around the portal area. CONCLUSION Compound Qizhu Tablets does have protective effects on the model rats with chronic liver injury and hepatic fibrosis, which is probably achieved thorough decreasing the inflammation.
