脓毒症大鼠肠道免疫功能损害的实验研究

         

摘要

目的 研究脓毒症大鼠肠道免疫功能的变化.方法 雄性SD大鼠20只,按简单随机法分为假伤组(10只)和脓毒症组(10只).脓毒症组大鼠予盲肠结扎穿刺造成脓毒症,假伤组大鼠除了不结扎和穿刺盲肠,其余处理同脓毒症组.于伤后48 h处死全部大鼠,取全段小肠.计数肠集合淋巴结(Peyer's patches,简称PP结)个数以及PP结淋巴细胞总数,用流式细胞仪检测PP结T和B细胞变化,采用ELISA检测肠黏液免疫球蛋白A(IgA)的变化.结果 与假伤组相比,脓毒症组大鼠PP结淋巴细胞总数显著减少、T和B细胞比例及绝对数量明显减少、肠黏液IgA相应减少(均P<0.05).结论 脓毒症导致大鼠肠道黏膜免疫功能抑制.%Objective To investigate the influence of sepsis on intestinal mucosal immune system. Methods Twenty SD rats were randomly divided into two groups, with 10 rats in each group. Experimental sepsis was induced by cecal ligation and puncture (CLP) in sepsis group. Except for CLP, the sham injury group was given the same treatment as sepsis group. All rats were sacrificed at 48 hours after CLP and entire intestines were collected. The number of intestinal Peyer's patches (PP) and total PP cell yields were counted. The percentages of B and T cells in PP were measured by flow cytometry. The levels of intestinal immunoglobulin A (IgA) were assayed by ELISA. Results Compared with sham injury group, total lymphocyte number in PP, percentages and numbers of T and B cells, and levels of intestinal IgA significantly decreased in sepsis group(P<0. 05). Conclusion Sepsis induced by CLP leads to intestinal mucosal immune suppression in septic rats.
