核不均一性核糖核蛋白H2(hnRNPH2)对单纯疱疹病毒(HSVI)复制影响

     

摘要

本文研究了单纯疱疹病毒(HSVI)在感染与复制过程中影响宿主细胞RNA转录,蛋白表达,进而为病毒的潜伏和持续感染创造必要条件.采用基于2-DE分析,人正常肝细胞L-02细胞核不均一性核糖核蛋白H2(heterogeneous nutlear ribonucleoprotein H2,hnRNPH2)在感染HSVI前后存在表达差异,构建pcDNA-hnRNPH2真核表达载体及hnRNPH2稳定转染的293T细胞系的方法,研究在核不均一性核糖核蛋白H2(hnRNPH2)稳定过表达后对HSVI病毒复制周期的影响.结果表明,病毒滴度和生长曲线显示,hnRNPH2可以显著影响病毒复制周期.过表达hnRNPh2的细胞系与对照相比,其病毒滴度明显下降,hnRNPH2在HSVI病毒复制中起到比较明显的抑制作用,其作用机理还有待进一步研究.%Herpes simplex virus I (nsv I ) regulating the pathway modify in host cell is a very systematic and complicate system. of transcription and translation By the analysis of 2-DE, the heterogeneous nuclear ribonucleoprotein H2 in human fetal liver cell represent distinction after the HSV I infection.
