生防真菌HPFJ3诱导小麦对全蚀病的抗性及其机理研究

     

摘要

为了明确生防真菌HPFJ3引入小麦根际对小麦植株体内微生物菌群的影响,对小麦全蚀病抗性的作用,以及对抗病性标志基因PR-1、PR-2、PR-5的激活作用,进一步了解小麦抗病性变化与基因表达变化之间的相关性。通过组织分离和稀释涂布平板分离法,分析生防真菌HPFJ3对小麦植株体内微生物菌群的影响;利用盆栽接种试验进行抗性测定;采用实时定量PCR技术检测小麦茎组织中PR-1、PR-2、PR-5基因的表达变化。结果表明:在小麦根际引入生防真菌HPFJ3,能显著增加小麦植株中的真菌和放线菌数量,而使体内细菌数量有一定程度的减少;明显提高小麦植株对全蚀病的抗性,其病情指数在整个生长时期均处于较低水平;生防真菌HPFJ3处理能诱导小麦植株中的PR-1、PR-2和PR-5基因表达水平增强,其3种抗性基因的表达均在小麦返青期达到了最大。生防真菌HPFJ3诱导的小麦抗病性和其对PR基因的诱导表达变化之间存在较好的一致性,为揭示生防真菌HPFJ3生防机理提供了科学依据。%This study aims to dertermine the effectes of inoculating HPFJ3 into wheat rhizospheres on the the number of endophytes of wheat, the wheat’s HPFJ3-induced resistance to take-all disease of wheat and the activation of the disease-resistance marker genes (PR-1, PR-2 and PR-5), so as to explicit the relationship between the induced resistance and the gene expression. The effect of inoculating HPFJ3 into wheat rhizospheres on the number of endophytea of wheat was studied by tissue separation and dilution plate separation methods,the wheat’s HPFJ3-induced resistance to take-all disease of wheat was assessed by pot experiments,and the real-time quantitative RT-PCR was used to detect the expressions of PR-1,PR-2 and PR-5. The results indicated: the inoculating HPFJ3 into wheat rhizospheres could significantly increase the number of fungi and actinomycetes in wheat plant and reduce the number of bacteria; the application of the strain HPFJ3 enhanced wheat’s resitance to take-all disease in the five periods and activated the expression of PR-1,PR-2 and PR-5, with the exppression of three resistant genes reaching their top value during the period of seedling-establishment;and the induced wheat resistance to take-all disease was positively associated with the induced expression of PR-1,PR-2 and PR-5. The study disclosed the bio-control mechanism of antagonistic fungi HPFJ3.
