PDGF-D对人胃癌细胞株SGC7901的作用

     

摘要

Aim:To investigate the expression and significance of PDGF-D and PDGFRβmRNA in human gastric cancer cell line SGC7901. Methods: The reverse transcription- polymerase chain reaction (RT-PCR) technique was used to examine mRNA expression of PDGF-D and PDGFRβ in human gastric cancer cell line SGC7901. SGC7901 were exposed to different concentration of PDGF-D(0,20,50,100, 500,2 500, 25 000,250 000 pg/mL) and the growth curve was drawn by MTT. The flow cytometer (FCM) was used to analyze the distribution of cells cycle. Results: PDGF-D and PDGFRβmRNA were expressed in human gastric cancer cell line SGC7901. PDGF-D enhanced SGC7901 proliferation (P <0. 05). The number of G0/G1 -phase cells decreased significantly compared with those of the controls while S-phase cells increased (P < 0. 05 ). Conclusion:PDGF-D may play a crucial role in carcinogenesis and development of gastric cancer. It can be used as a new therapeutic target of gastric cancer in clinioal practice.%目的:探讨血小板衍生生长因子-D(PDGF-D)及其受体PDGFRβmRNA在人胃癌细胞株SGC7901中的表达及其意义.方法:用RT-PCR方法检测人胃癌细胞株SGC7901中PDGF-D及PDGFRβmRNA的表达,将人重组PDGF-D蛋白分别以0、20、50、100、500、2 500、25 000、250 000 pg/mL浓度加入人胃癌细胞株SCG7901中,采用四甲基偶氮唑蓝比色法(MTT法)检测胃癌细胞株SCG7901的增殖情况并画出生长曲线;采用流式细胞仪检测细胞周期变化.结果:PDGF-D及PDGFRβ mRNA在胃癌细胞株中表达.MTT检测发现外源性PDGF-D蛋白可以促进SGC7901细胞增殖(P<0.05).细胞周期分析显示外源性PDGF-D蛋白干预细胞后实验组G0/G1期细胞比例下降,S期细胞比例上升(P<0.05).结论:PDGF-D在胃癌的发生、发展中可能起着重要的作用.PDGF-D可能成为治疗胃癌的新的靶点.
