大孔吸附树脂法提取急性子总皂苷的方法

     

摘要

Four kinds of main hosenkosides named as hosenkosides K, A, C,B were used as the indexes of total hosenkosides extracted from the seeds of Impatiens balsamina L. The effects of the density, pH and the rate of the flow of the solution on the adsorption properties of the resin were investigated through the orthogonal test, establishing the adsorption conditions. The results show that the solutions were combined after filtration and evaporated to the 6 times the material weight in vacuum. The solution at pH 6.0 was passed through the AB-8 macroporous resin at a flow rate of 3 volumes of the resin in 1 h, then eluted by 80% ethanol. This way could collect the total hosenkosides, in which the content of the sum of four hosenkosides K, A, C, B was in the highest point.%以急性子中4种主要皂苷类化合物凤仙萜四醇苷K,A,C,B质量分数之和为考察指标,通过正交试验考察了药液的质量浓度、pH值、流速、乙醇洗脱液的体积分数等影响AB-8大孔吸附树脂吸附性能的主要因素,确定了最优吸附条件.确定最优的提取方法为:将急性子用体积分数为70%的乙醇回流提取,提取液浓缩至药材质量的6倍即得药液,调节其pH值约为6.0,每小时以3倍树脂体积的流速通过AB-8大孔吸附树脂柱,用体积分数为80%的乙醇洗脱,此时洗脱物中四种凤仙萜四醇苷K,A,C,B质量浓度之和最高.
