人参糖蛋白对小鼠行为学和海马内单胺类神经递质的影响

     

摘要

试验研究了人参糖蛋白(Glycoprotein from Panax ginseng,PGG)对行为绝望模型小鼠海马内单胺类神经递质的影响.将50只小鼠(昆明种)随机分为5组,采用悬尾试验和强迫游泳试验考察PGG对绝望模型小鼠行为学的影响;利血平诱导试验考察PGG对小鼠体温和眼睑的影响;采用高效液相色谱仪结合电化学检测其考察PGG对小鼠海马内5-羟色胺(5-hydroxy tryptamine,5-HT)和5-羟吲哚乙酸(5-hydroxyindole acetic acid,5-HIAA)含量的影响.试验结果表明:与生理盐水组相比,PGG中、高剂量可以明显减少悬尾和强迫游泳的不动时间(P<0.01),逆转利血平诱导的体温降低和眼睑下垂(P<0.05),并提高海马内5-HT及其代谢产物的水平(P<0.05).这说明PGG对行为绝望模型小鼠具有保护作用,可能是通过升高海马内单胺类递质的含量实现的.%This study investigated the effects of glycoprotein from Panax ginseng (PGG) on behavioral changes and hippocampal monoamine neurotransmitters levels of behavioral despair models in mice.Fifty Kunming mice were randomly divided into 5 groups.Tail suspension test and forced swimming test were carried out to explore the effects of PGG on behavioral changes.The effects of PGG on reserpine-induced hypothermia and ptosis were also investigated.Content of 5-HT and 5-HIAA in hippocampus of mice was detected by using HPLC with electrochemical detection.The results showed that compared with the saline group,middle and high doses of PGG can reduce immobility time in tail suspension and forced swimming,reverse reserpine-induced hypothermia and ptosis,and increase hippocampus 5-HT and its metabolite levels.The experimental results show the protective effects of PGG on depression model mice,possibly via increasing the content of monoamine neurotransmitters in the hippocampus.
