漆酶对纤维素酶催化作用的影响

         

摘要

In order to investigate the effect of laccase on catalytic efficiency of cellulase,a comparative analysis of sugar yield from cellulosic hydrolysate of filter paper and corn stover using different addition level of the pure laccase(Sigma) was performed in this study.The results indicated that the addition of lactase had no significant effect on the sugar yield in the filter paper hydrolysis system which contained cellulase.However,the addition of lactase could decrease the sugar yield (from 30.40% to 23.87%) obviously in the corn stover hydrolysis system (preincubating laccase and corn stover together at 37℃ for 24 h).A negative correlation between the sugar yield and the addition of lactase in the corn stover hydrolysis system was also observed.Hence,laccase had played an inhibitory role in cellulase catalysis in the corn stover hydrolysis system.%为探究漆酶对纤维素酶催化作用的影响,以Sigma纯漆酶为供试酶,对比分析不同添加量的漆酶在纤维素酶降解滤纸和降解汽爆玉米秸秆时糖得率的变化.结果表明,在纤维素酶降解滤纸的体系中,加入漆酶对于糖得率无明显影响;而在纤维素酶降解玉米秸秆的体系中,加入漆酶会使玉米秸秆的糖得率降低,在漆酶与秸秆底物37℃保温24 h条件下,最大可使糖得率由30.40%降低至23.87%,且糖得率的降低与漆酶添加量呈负相关关系.研究证实漆酶的加入对纤维素酶水解玉米秸秆具有抑制作用.
