西伯利亚鸢尾茎尖离体培养快繁技术研究

         

摘要

以西伯利亚鸢尾蓝国王的茎尖为外植体,研究不同培养基对其愈伤组织诱导、不定芽分化、增殖和生根的影响.结果表明:茎尖愈伤组织诱导的适宜培养基为MS+KT 1.0 mg/L+2,4-D1.0 mg/L,诱导率为62.37%;培养基MS+6-BA 1.0 mg/L+NAA 0.2 mg/L有利于愈伤组织的分化,不定芽分化率为60.24%;采取单芽纵切的方式在培养基MS+6-BA 2.0 mg/L+NAA 0.2mg/L中培养芽增殖情况良好,平均增殖倍数为6.8;不定芽在培养基1/2MS+NAA 0.5 mg/L+活性炭(AC)0.5 g/L中生根率为97.64%.幼苗移栽蛭石中培养15 d后成活率达96%.%The shoot tips of Iris sibirica L.were used as explants to develop a protocol for callus induction,shoot formation,rapid propagation and rooting.The results showed that suitable medium for callus induction was MS medium supplemented with 1.0 mg/L KT and 1.0 mg/L 2,4-D,where callus induction rate reached 62.37 %.MS medium supplemented with 1.0 mg/L 6-BA and 0.2 mg/L NAA was in favor of the differentiation of shoots from the callus,with the differentiation rate of 60.24%.Single bud with base slitting was subcultured on MS medium supplemented with 2.0 mg/L 6-BA and 0.2 mg/L NAA and obtained a higher proliferation rate of 6.8.The suitable rooting medium was 1/2MS medium supplemented with 0.5 mg/L NAA and 0.5 g/L activated carbon,on which rooting rate reached 97.64 %.Vermiculite was suitable for the transplanting,in which the surviving rate was more than 96%.
