牡丹品种岛锦试管苗与大田苗生理指标差异研究

         

摘要

Root activity,contents of chlorophyll,soluble protein,soluble sugar,and activities of peroxidase ( POD) ,polyphenol oxidase( PPO) ,superoxide dismutase( SOD) ,catalase( CAT) activities were assessed in leaves and roots of tree peony cultivar Daojin grown in vitro and in field. And the differences between the test-tube plantlets and field seedlings were compared. The results showed that there were great differences in physiological indexes between the test-tube plantlets and field seedlings of tree peony, root activity,contents of chlorophyll,soluble protein and activities of PPO,SOD of the test-tube plantlets were lower than that of field seedlings,root activity of field seedlings was 689 μg/(g·h),and that of the test-tube plantlets was only 141 μg/( g·h) ,but the POD activity of the plantlets was higher than that of field seedings,POD ac-tivities in leaves and roots of test-tube plantlets were 432. 37 U/(g·min),43. 89 U/(g·min) respective-ly,and which of field seedlings were 1. 76 U/(g·min) and 1. 61 U/(g·min) respectively. And the simi-lar rule did not appear in soluble sugar content and CAT activity. The survival rate of transplanting testtube plantlets was low,which may be related with the low root activity,low contents of chlorophyll and soluble protein,low activities of PPO and SOD,and high POD activities.%为了探明牡丹试管苗与大田苗生理指标的差异,揭示影响牡丹试管苗移栽成活的可能因素,以牡丹品种岛锦试管苗和大田苗的叶片及根系作为研究材料,分析了两者根系活力、叶绿素含量、可溶性蛋白含量、可溶性糖含量以及过氧化物酶( POD )、多酚氧化酶( PPO )、超氧化物歧化酶( SOD)和过氧化氢酶( CAT)活性的差异。结果表明:牡丹试管苗与大田苗的生理指标存在很大差异,试管苗的根系活力、叶绿素含量、可溶性蛋白含量以及PPO、SOD活性等指标均比大田苗低,大田苗根系活力为689μg/( g·h),而试管苗仅为141μg/( g·h),但试管苗的POD活性明显比大田苗高,试管苗叶片和根系的POD活性分别为432.37 U/(g·min)、43.89 U/(g·min),而大田苗叶片和根系的POD活性仅为1.76 U/(g·min)和1.61 U/(g·min)。可溶性糖含量和CAT活性未表现出类似的规律。牡丹试管苗移栽成活率低可能与其根系活力、叶绿素含量、可溶性蛋白含量以及PPO、SOD活性较低,POD活性较高有关。
