理冲生髓饮中多糖及水溶性蛋白质的质量分数测定

         

摘要

建立理冲生髓饮中多糖及水溶性蛋白质质量分数测定的方法.测定理冲生髓饮多糖对葡萄糖的换算因子,利用苯酚-硫酸法测定多糖的质量分数;采用考马斯亮蓝G-250染色法测定理冲生髓饮中可溶性蛋白质的质量分数.多糖标准曲线方程为A =12.839 C-0.0085(r =0.9993),蛋白质标准曲线方程为A =1.271 C-0.0331(r =0.9992),两者分别在0.01~0.1、0.1~1.0 g/L有良好的线性关系.测得理冲生髓饮中多糖质量分数为8.53%,水溶性蛋白质质量分数为0.35%.该两种方法操作简便、灵敏、快速、准确,重复性好,可用于理冲生髓饮中多糖、水溶性蛋白质的质量分数测定.%To establish a method for the determination of polysaccharide and water -soluble protein in Lichong Shengsui Drink ( LCSSD) , the conversion factor of LCSSD polysaccharide to glucose was obtained with refined polysaccharide , and the content of LCSSD polysaccha-ride was determined by using phenol -sulfuric method .The content of protein was deter-mined by using the dying method with Coomassie brilliant blue G -250.The polysaccharide standard curve equation was A =12.839 C -0.008 5 (r =0.999 3) and protein standard curve equation was A =1.271 0 C -0.033 1 (r =0.999 2).Respectively the linear range was 0.01~0.1 g/L and 0.1~1.0 g/L.The content of polysaccharide was 8.53%, and the content of protein was 0.35%.The method is simple, fast, accurate and reproduci-ble, and it should be helpful for the determination of polysaccharide and water -soluble pro-teins in LCSSD .
