补肾活血方对膝骨关节炎大鼠关节软骨HIRA、ASF1a表达的影响

摘要

[目的]探讨补肾活血方对膝骨关节炎大鼠关节软骨衰老信号分子HIRA、ASF1a表达的影响.[方法]选用雌性SD大鼠,随机分为假手术组、模型组和中药组；采用切除双侧卵巢并切断右侧膝交叉韧带法复制骨关节炎动物模型,术后第4周开始灌服补肾活血中药,模型组、假手术组灌服等量的生理盐水,分别于术后8、12、16周取材,采用骨关节炎软骨病理变化评价系统(OARSI)评价关节软骨的病变,运用western blot法检测H1RA、ASF1a表达水平.[结果]术后8、12、16周,模型组OARSI评分较假手术组显著升高,差异有显著性意义(P＜0.01),中药组OARSI评分较模型组显著降低,差异有显著意义(P＜0.05)；HIRA、ASF1a在模型组中表达明显增强,补肾活血中药能下调HIRA、ASF1a的表达.[结论]补肾活血方可通过下调HIRA、ASF1a表达保护关节软骨.%Objective To observe the influence of Bushen Huoxue Recipe (BHR), which has the actions of tonifying kidney and promoting blood circulation, on senescence-associated factors ASFla and HIRA expression in articular cartilage of rats with knee osteoarthritis. Methods Female SD rats were randomized into three groups: sham-operation group, model group and BHR group . By removing bilateral ovaries and cutting off cruciate ligaments of the right knee, we established osteoarthritis animal model. Four weeks after the operation, rats in BHR group were given gastric gavage of BHR, and the model group and sham-operation group were given the same volume of normal saline. Osteoarthritis cartilage pathological changes evaluation system ( OARSI) was used to evaluate the pathological features of articular cartilage, and Western blotting method was used for the examination of ASFla and HIRA expression 8, 12 and 16 weeks after the operation. Results OARSI score was increased in the model group (P < 0. 01 compared with that in the sham-operation group) , while was decreased in BHR group (P < 0.05 compared with that in the model group) 8, 12 and 16 weeks after the operation. ASFla and HIRA expression was up-regulated in the model group compared with that in the sham-operation group, and was down-regulated in BHR group compared with that in the model group. Conclusion BHR has protective effect on articular cartilage of rats with knee osteoarthritis through down-regulating cartilaginous ASFla and HIRA expression.
