针药结合对围绝经期模型大鼠性激素及胰岛素样生长因子的影响

     

摘要

[目的]探讨电针加雌激素(E2)治疗对围绝经期模型大鼠性激素和胰岛素样生长因子(IGF)的影响.[方法]选用雌性未孕Wistar大鼠随机分为正常组、模型组、假手术组、雌激素组(剂量为0.04 mg·kg-1·d-1),电针组(电针肾俞、太溪、足三里、三阴交),电针加雌激素组(电针方法同电针组,雌激素剂量减半),采用双侧卵巢摘除术复制围绝经期模型,造模成功后治疗3个疗程,检测各组大鼠性激素和IGF-1、IGF-2水平的变化.[结果]模型组大鼠性激素水平出现明显变化,E2水平下降,黄体生成素(LH)、卵泡刺激素(FSH)水平升高,IGF-1含量下降,与正常组比较有显著性差异(P<0.05或P<0.01);治疗后,雌激素组、电针组、电针加雌激素组E2水平升高,LH、FSH水平降低,IGF-1含量升高,与模型组比较有显著性差异(P<0.05或P<0.01),其中电针加雌激素作用最为明显.各组IGF-2水平无显著变化(P>0.05).[结论]在减少雌激素用量的情况下,电针配合雌激素治疗对围绝经期模型大鼠性激素及IGF水平具有良好的调节作用.%Objective To investigate the influences of electroacupuncture (EA) combined with estrogen (E2) on sexual hormones and insulin-like growth factor (IGF) in peri-menopausal rats. Methods Female unpregnant Wistar rats were randomly divided into normal group, model group, sham operation group, estrogen group (in the dosage of 0. 04 mg ? kg-1 ? d-1 ) , EA group ( EA on points of Shenshu, Taixi, Zusanli and Sanyinjiao), and combination group ( EA combined with estrogen in half of the dosage). The model was established by bilateral ovariectomy. After the modeling, the rats were given corresponding treatment for 3 courses. The serum contents of E2, follicle-stimulating hormone (FSH) , luteinizing hormone (LH), IGF-1 and IGF-2 were measured. Results The contents of E2 and IGF-1 significantly decreased, and FSH and LH increased in the model group ( P < 0. 05 or P < 0. 01 compared with those in the normal group). After treatment, the contents of E2 and IGF-1 significantly increased, FSH and LH decreased obviously in estrogen group, EA group and combination group (P < 0. 05 or P <0. 01 compared with those in the model group) , and the effect in the combination group was the strongest. The changes of IGF-2 were not obvious in all groups (P > 0.05). Conclusion EA combined with dosage-reduced estrogen exerts satisfactory regulation on sexual hormones and insulin-like growth factor in peri-menopausal rats.
