高效液相色谱法测定复方伯氨喹片中青蒿素与磷酸伯氨喹的含量

     

摘要

Objective To establish an HPLC method for quantitative determination of artemisinin and primaquine phosphate components in Compound Primaquine Tablets. Methods For artemisinin, the liquid chromatography was carried out on C18 column. The mobile phase consisted of acetonitrile-water ( volume ratio being 68: 32). The flow rate was 0.8 mL/min, detection wavelength was 216 nm and the column temperature was 251. For primaquine phosphate, the liquid chromatography was carried out also on Clg column. The mobile phase consisted of acetonitrile -0. 25% diethylamine ( volume ratio being 30:70) , adjusted to pH value being 4. 0 with phosphoric acid. The flow rate was 0. 8 mL/min, detection wavelength was 263nm and the column temperature was 251. Results For artemisinin, the calibration curve was linear in the range of 8 ~ 40 μg (r = 0. 999 992), and the average recovery (N = 9 ) was 99. 8%. For primaquine phosphate, the calibration curve was linear in the range of 0. 032-1. 120 μg (r = 0. 999 969), and the average recovery (N = 9) was 99. 9%. Conclusion The method is accurate, convenient, rapid and reliable, and is suitable for the determination of artemisinin and primaquine phosphate in Compound Primaquine Tablets.%[目的]建立以高效液相色谱法测定复方伯氨喹片中青蒿素与磷酸伯氨喹含量的方法.[方法]青蒿素采用C18柱,以乙腈-水(体积比为68:32)为流动相,流速:0.8 mL/min,检测波长:216 nm,柱温:25℃;磷酸伯氨喹采用C18柱,以乙腈-2.5 g/L二乙胺溶液(磷酸调pH至4.0,体积比为30:70)为流动相,流速:0.8 mL/min,检测波长:263 nm,柱温:25℃.[结果]青蒿素进样量在8~40 μg范围内与峰面积呈良好的线性关系,r=0.999 992,平均回收率为99.8%;磷酸伯氨喹进样量在0.032~1.120 μg范围内与峰面积呈良好的线性关系,r=0.999 969,平均回收率为99.9%.[结论]本研究方法结果准确可靠,简便快捷,适用于复方伯氨喹片中青蒿素与磷酸伯氨喹含量的测定.
