九节木离体培养的研究

     

摘要

[目的]采用植物组织培养技术进行植株再生和快速繁殖,为建立九节木的快速繁殖再生体系奠定基础.[方法]以九节木的幼嫩果实和叶片为外植体,以MS为基本培养基,对九节木外植体进行表面消毒,筛选适宜的外植体消毒时间;研究MS及MS辅以活性炭对外植体生长的影响;通过正交试验设计,研究不同种类及浓度配比激素对丛生芽增殖的影响;对获得的试管苗炼苗与移栽.[结果]以果实为外植体较为适宜;采用1 g/L HgCl2对九节木外植体表面消毒15 min效果较好;添加活性炭有利于外植体生长;丛生芽增殖效果较好的培养基为MS +2.0 mg/L 6-BA +0.5 mg/L NAA +0.5 mg/L IBA;移栽到室内1周,成活率达80.0%.[结论]建立了九节木离体快速繁殖体系.%Objective To introduce plant tissue culture technology for the regeneration and rapid propagation of Psychotria rubra ( Lour. ) Poir., thus to lay the foundation for the regeneration and rapid propagation system of Psychotria rubra ( Lour. ) Poir. Methods With the leaves and the young fruits of Psychotria rubra ( Lour. ) Poir. As the explants, and with MS as the basic medium, we optimized the explants and time for surface sterilization, and observed the effect of MS as well MS supplemented with activated carbon on the growth of the explants. By orthogonal design, we observed the effect of various kinds of hormones and various matching concentrations of hormones on the proliferation of clustered buds. And then the obtained test-tube plantlet was used for acclimatization and transplantation. Results Fruits were the optimal explants of Psychotria rubra, and disinfection with 1 g/L HgCl2 for 15 min was the best. Adding activated carbon was benefit for explain growth. The optimal basic medium for promoting clustered buds proliferation was MS +6-BA 2. 0 mg/L + NAA 0. 5mg/L + IBA 0. 5mg/L. The test-tube plantlet got a survival rate of 80% after indoor transplanting for one week. Conclusion In-vitro culture system of Psychotria rubra ( Lour. ) Poir. Has been established .
