Objective:To investigate the impacts of Numb protein on epithelial-mesenchymal transition ( EMT) in esophageal squamous cell carcinoma ( ESCC ) . Methods: The siRNA technique was employed to down-regulate endogenous Numb expression of KYSE150 cells. Wound healing assay and Boyden Chamber assay were used to investigate the influence of Numb protein on the mobility of KYSE150. Western blotting was adopted to explore the impacts of Numb on the E-cadherin and vimentin expression. Results:Numb-siRNA resulted in a marked reduction in Numb expression in KYSE150 cells. The scratch wound healing assay and Boyden chamber assay demonstrated that the motility and invasiveness of KYSE150 cells were, following induction with 50 μg�L-1 epidermal growth factor, dramatically enhanced. The knockdown of Numb protein was associated with a notable decrease in E-cadherin and up-regulation of Vimentin expression,which were not observed in the control group. Conclusion:Numb protein suppresses the mobility of ESCC via inhibition of EMT.%目的:探讨Numb蛋白的表达对食管鳞癌细胞间质转化过程( EMT)过程的影响。方法:使用siRNA干扰技术下调KYSE150细胞内源性Numb的表达。随后使用划痕实验和Transwell观察Numb对KYSE150细胞迁移能力的影响。 Western blotting检测E-cadherin和Vimentin蛋白的表达。结果:Numb-siRNA能有效地减低 KYSE150内源性 Numb 的表达。划痕实验和 Transwell 实验显示,干扰了内源性Numb表达的KYSE150细胞的运动迁移能力明显增强。在50μg/L的EGF诱导下,干扰了Numb表达的KYSE150细胞的E-cadherin明显下降,Vimentin表达明显增高,而阴性对照组的E-cadherin和Vimentin变化均不明显。结论:Numb能通过抑制食管鳞癌细胞的EMT过程从而抑制细胞的迁移能力。
