Caveolin-1蛋白对肝细胞癌ERK信号转导通路的影响

         

摘要

Objective:To investigate the expression and significance of caveolin-1 and ERK-1 protein in hepa-tocellelar carcinoma (HCC). Methods: The expression of caveolin-1 and ERK-1 protein was detected by immunohistochemistry in 10 cases of normal liver tissue, and 30 cases of liver cancers and paracancer liver tissue. Results: ① The expression rate of caveolin-1 protein in normal liver tissue, paracancer liver and liver cancers tissue was 90%, 13. 33% and 10% ( P <0. 05), respectively. The expression rate of ERK-1 protein was 30% , 56. 67% and 83. 33% ( P <0. 05) , respectively. ② The expression of caveolin-1 protein was negatively correlated with the expression of ERK-1 protein ( r = - 0. 447, P <0. 05). Conclusion: Caveolin-1 protein may have inhibitory effect in HCC of the occurrence and development process. Low expression of caveolin-1 protein may activate ERK-1 protein, and ERK-1 signal transduction path-ways lose normal coordinate the balance which is the important factor that could lead to HCC of the occurrence.%目的:研究小窝蛋白-1(Caveolin-1)和细胞外信号调节激酶-1(ERK-1)蛋白在肝细胞癌中的表达及意义.方法:采用免疫组织化学SP法检测30例肝细胞癌组织、相应癌旁肝组织及10例正常肝组织中Caveolin-1蛋白和ERK-1蛋白的表达.结果:①Caveolin-1蛋白在正常肝组织、癌旁肝组织、肝细胞癌组织中的阳性表达率分别为90%、13.33%、10%.ERK-1蛋白在正常肝组织、癌旁肝组织、肝细胞癌组织中的阳性表达率分别为30%、56.67%、83.33%.Caveolin-1蛋白和ERK-1蛋白在不同组织间差异均有统计学意义(P<0.05);②Caveolin-1蛋白与ERK-1蛋白的表达在肝细胞癌中呈负相关(r=-0.447,P<0.05).结论:①Caveolin-1蛋白可能在肝细胞癌的形成过程中起抑制作用.②Caveolin-1蛋白表达下调从而可能激活ERK-1蛋白,使ERK细胞信号转导通路失去正常的协调平衡可能是导致肿瘤发生的重要因素.
