1株红树林可口革囊星虫共附生放线菌抑制COX-2活性成分的研究

             

摘要

Objective To isolate and screen Phascolosoma esculenta symbiotic actinomycetes from mangrove for COX-2 inhibitory activity, and study secondary metabolites from the active strain PE05. Methods The symbiotic actinomycetes were isolated using dilution-spreading-plate method. The COX-2 inhibitory activities of fermentation products from all of strains were evaluated by COX inhibitor screening assay kit. PE05 was confirmed as the target strain and isolated by various column chromatography and recrystallization. Moreover, the structures of pure compounds were identified by spectroscopic methods. Results Eleven symbiotic actinomycetes were isolated from Phascolosoma esculenta, and six of them showed COX-2 inhibitory activities. Five compounds were obtained from PE05, which were identified as vanillic acid (1), 4-hydroxybenzoie acid (2), phenylethyl acid (3), nicotinic acid (4), cyclo- (Pro-Leu) (5). Conclusion PE05 of Phascolosoma esculenta could be a suitable candidate as a potential COX-2 inhibitor producer, which will provide a new strategy for COX-2 inhibitor discovery.%目的 从红树林可口革囊星虫表皮中分离共附生放线菌并进行抑制COX-2活性筛选,对其中1株具有较强活性菌株PE05的活性成分进行研究.方法 采用稀释涂布法对红树林可口革囊星虫共附生放线菌进行分离,通过COX抑制活性试剂盒筛选菌株发酵产物抑制COX-2的活性.结合生物活性和薄层色谱检测,以PE05作为目标菌株,使用多种柱层析、重结晶等方法对发酵产物粗提物进行分离纯化,并通过现代波谱学技术鉴定化学结构.结果 从红树林可口革囊星虫表皮中分离得到11株共附生放线菌,对其发酵产物进行活性筛选显示其中6株具有较好的抑制COX-2活性.从高活性菌株PE05中分离得到5个化合物,分别鉴定为:香草酸(1)、对羟基苯甲酸(2)、苯乙酸(3)、烟酸(4)、环(脯-亮)二肽(5).结论 从红树林可口革囊星虫共附生放线菌中发现PE05能产生良好COX-2抑制活性的代谢产物,该结果为从红树林生态系统中发掘新型的COX-2抑制剂提供了一种新的策略.
