远志皂苷调节细胞自噬抗Aβ25-35诱导的SH-5Y5 Y细胞凋亡的研究

         

摘要

目的 从细胞自噬角度探讨远志皂苷(tenuifolin,Ten)抑制Aβ25-35诱导SH-SY5Y细胞凋亡的作用及分子机制.方法 建立Aβ25-35诱导的SH-SY5Y细胞损伤模型,通过MTT法和流式细胞术测定细胞活力和凋亡率,单丹磺酰尸胺(MDC)染色检测自噬小泡的变化,Western blot检测LC3-Ⅱ/LC3-Ⅰ蛋白表达水平的变化.结果 Ten抑制Aβ25-35诱导SH-SY5Y细胞活力降低及凋亡率增加,减少Aβ25-35诱导的自噬体增加,显著降低自噬蛋白LC3-Ⅱ/LC3-Ⅰ的表达水平.结论 Ten通过调节Aβ25-35诱导的细胞自噬,从而抑制Aβ25-35诱导的SH-SY5Y细胞凋亡,发挥神经保护作用.%Objective To investigate the effect of tenuifolin ( Ten) against Aβ25-35-induced SH-SY5Y cell apoptosis and its molecular mechanism from the perspective of cell autophagy. Methods Aβ25-35-induced SH-SY5Y cell damage model was established. Cell viability and apoptosis were determined by MTT and flow cytometry. The changes of autophagic vacuoles in SH-SY5Y cells were observed by MDC staining, and microtubule-associated protein light chain 3 ( LC3) was detected by Western blot. Results Ten inhibited the cell viability decrease and apoptosis of SH-SY5Y cells induced by Aβ25-35. Ten reduced Aβ25-35-induced cell autophagy and the expression of LC3-Ⅱ/LC3-Ⅰ protein. Conclusion Ten can inhibit Aβ25-35-induced SH-SY5Y cell apoptosis by regulation of cell autophagy, and thus plays a neuroprotective role.
