萝卜硫素下调Traf6/NF-κB信号通路抑制肾癌细胞增殖的作用研究

         

摘要

Objective To investigate the effect and mechanism of sulforaphane on the proliferation of kidney cancer RC-2 cells. Methods RC-2 cells and Traf6-overexpressed RC-2 cells were cultured in vitro. The cell proliferation was measured by CCK8 assay. The protein and mRNA expression of Traf6 and NF-κB were detected by western blot and RT-PCR. Results Treatment with over 10 μmol/L sulforaphane inhibited RC-2 cell proliferation in a dose-dependent way. The over-expression of Traf6 promoted the proliferation of RC-2 cells. Sulforaphane ( 20 μmol/L) inhibited downregulation of NF-κB induced by Traf6 expression. Conclusion Sulforaphane could suppress the proliferation of RC-2 cells via down-regulation of Traf6/NF-κB signaling.%目的 探讨萝卜硫素对肾癌细胞(RC-2)增殖的影响及其机制.方法 体外培养RC-2和Traf6过表达的RC-2细胞,不同浓度萝卜硫素处理对数生长期的细胞株,采用CCK8试剂盒检测萝卜硫素对RC-2细胞和过表达Traf6基因RC-2细胞增殖的抑制作用;免疫印迹(Western blot)和反转录聚合酶链反应(RT-PCR)方法检测RC-2细胞中Traf6和NF-κB蛋白及mRNA的表达水平.结果 萝卜硫素的浓度大于10μmol/L时可抑制RC-2细胞的增殖,并随着浓度增加抑制率随之增强;Traf6的过表达可以促进RC-2细胞的增殖;RT-PCR和Western blot实验结果表明20μmol/L的萝卜硫素可以抑制Traf6的表达而引起NF-κB表达下调.结论 萝卜硫素可以抑制RC-2细胞的增殖,其机制可能与下调Traf6/NF-κB信号通路蛋白表达有关.
