一株降解纤维素嗜热真菌的筛选、鉴定及其产酶条件的优化

     

摘要

Using sodium carboxymethyl cellulose as sole carbon resource, a thermophile fungi YT-40-1 degrading cellulose was isolated from hot spring of Yongtai County in Fuzhou. Based on rDNA sequence analysis, the strain was identified as Candida parapsi-losis. The enzyme yield of strain YT-40-1 reached its peak after culturing 72 h under the conditions as follows; initial pH was 4.0, temperature was 50 ℃ , inoculum was 10% , rotation speed was 200 r ? Min-1.%以羧甲基纤维素钠为唯一碳源,从福州永泰温泉分离到1株降解纤维素的嗜热菌YT-40-1,经过形态观察和ITS rD-NA序列比较分析,初步鉴定为近平滑假丝酵母(Candida parapsilosis).菌株YT-40-1在初始pH为4.0、温度为50℃、接种量为10%、摇床转速为200 r·min-1的条件下在培养基中培养72 h,总酶活力达到最大.
