肝细胞癌组织Bcl-2和Bax蛋白的表达(英文)

     

摘要

目的探讨原发性肝细胞癌(简称肝癌)细胞凋亡相关蛋白Bcl-2和Bax的表达及其意义. 方法采用免疫组化ABC法检测Bcl-2和Bax蛋白在66例人肝癌组织和24例人正常肝组织,以及3株人肝癌细胞系HCC-9204, SMMC-7721, HHCC和1株永生化的人正常肝细胞系QZG的表达情况,并分析肝癌组织的Bcl-2和Bax阳性率与其病理分级之间的关系. 结果 Bcl-2在正常肝组织的阳性率只有4.2%,而在肝癌组织为25.8%差异显著(P<0.05). Bax在正常肝组织的阳性率为70.8%,在肝癌组织为43.9%,差异显著(P<0.05). 肝癌组织的Bcl-2和Bax阳性率与其病理分级之间均未见明显的相关性(P>0.05). 在正常肝和肝癌组织中,Bcl-2阳性率均明显低于Bax(P<0.01, P<0.05). 在QZG细胞中未见明显的Bcl-2阳性信号,Bax阳性细胞计数率为61%. SMMC-7721细胞和HHCC细胞的Bcl-2阳性细胞计数率分别为14.3%和11.8%,Bax阳性细胞计数率分别为58.3%和47.1%. HCC-9204细胞中未见明显的Bcl-2阳性信号,Bax阳性细胞计数率为43.2%. 结论与正常肝组织相比,肝癌组织的Bcl-2蛋白低度升高,Bax蛋白下降. 正常肝和肝癌组织的Bcl-2水平均低于Bax. 肝癌和正常肝细胞系的Bcl-2蛋白水平均很低,可见中等程度Bax蛋白表达.%AIM To elucidate significance and expression of apoptosis-related proteins of Bcl-2 and Bax in primary hepatocellular carcinoma (HCC). METHODS Bcl-2 and Bax proteins were detected in 66 human HCC tissues, 24 human normal liver tissues, 3 human HCC cell lines (HCC-9204, SMMC-7721 and HHCC) and a human immortalized normal hepatocyte cell line QZG by immunohistochemical ABC method. The positive rates and their relationship with pathological grades of HCC tissues were further investigated. RESULTS The positive rate of Bcl-2 was only 4.2% in normal liver tissues, whereas 25.8% (17/66) in HCC (P<0.05). The positive rate of Bax in normal liver tissues was 70.8%, while that in HCC tissues was 43.9% (P<0.05). The positive rates of both Bcl-2 and Bax had no distinct correlation with pathological grades of HCC tissues (P>0.05). The positive rate of Bcl-2 as obviously lower than that of Bax in both normal liver tissues (P<0.01) and HCC tissues (P<0.05). There was no detectable Bcl-2 positive signal, and the counting rate of Bax positive cells was 61% in QZG cells. The counting rates of Bcl-2 positive cells were 14.3% and 11.8%, and those of Bax positive cells were 58.3% and 47.1% in SMMC-7721 cells and HHCC cells, respectively. There was no detectable Bcl-2 positive signal, and the counting rate of Bax positive cells was 43.2% in HCC-9204 cells. CONCLUSION Compared with that in normal liver tissues, the expression of Bcl-2 protein increases a little and Bax protein decreases in HCC tissues. Bcl-2 level is lower than Bax level in both normal liver and HCC tissues. The Bcl-2 protein level is very low and that of Bax is moderate in both HCC and normal hepatocyte cell lines.
