本研究旨在探讨中国汉族人群中人类白细胞抗原(HLA)DR和DQ基因连锁不平衡的分布规律,提高HLA配型的准确性.采用PCR-SSP基因分型方法,对1799例汉族受检者的DR和DQ基因进行了分型,对其基因的连锁不平衡进行了研究.结果发现,在1799例受检者中有29种不同于西方人种的新连锁组合,其中DR13-DQ5出现频数最高,达11次;DR11-DQ9出现8次,DR8-DQ8出现7次;DR9-DQ8和DR12-DQ6及DR14-DQ4各出现6次.所有29种新单倍型的连锁不平衡参数均为负,其连锁单倍型的发生频率低于理论频率,显示新发现的连锁不平衡单倍型都比较罕见,在人群中的分布频率较低.结论:本研究丰富了经典HLA-DR/DQ连锁组合,指出了该种组合分布的民族相关性,对提高HLA配型实验的准确性和工作效率具有重要意义.%This study was aimed to investigate the distribution feature of HLA-DR/DQ gene linkage disequilibrium in Chinese Han population and to improve the accuracy of HLA matching results. Genotyping of HLA-DR and HLA-DQ gene locus was performed using PCR-SSP typing in Chinese Han population receiving kidney transplantation. The results showed that there were 29 new linkage combinations in 1799 patients, in which DR13-DQ5, DR11-DQ8 and DR8-DQ8 were discovered for 11, 8 and 7 times respectively while DR9-DQ8 ,DR12-DQ6 and DR14-DQ4 were both discovered for 6 times. The linkage disequilibrium parameters of these haplotypes were negative, showing that these linkages were uncommon. It is concluded that this study not only enriches the classical HLA-DR/DQ linkage combinations, but also indicates the national relevance of combination distribution, and it has great importance in improving the accuracy of HLA matching experiments and reducing unnecessary repeated work.
