目的 研究美珍颗粒剂治疗溃疡性结肠炎的作用机制.方法 用2,4二硝基氯苯建立大鼠创伤性溃疡结肠炎模型,观察美珍颗粒剂对其结肠黏膜组织病理学、结肠损伤分数、红细胞超氧化物歧化酶(SOD)和结肠黏膜过氧化脂质(LPO)的影响.结果 造模大鼠经治疗,结肠黏膜光镜下组织学有明显改变,结肠损伤分数、大便乳酸含量降低,SOD升高,LPO降低,与模型组比较,有显著差异.美珍颗粒剂组治疗后核转录因子κB(NF-κB),白细胞介素(IL)-6,IL-1β因子表达进一步减弱,与SASP组相比有明显差异.结论 美珍颗粒剂发挥抗溃疡性结肠炎的机制之一可能与其减弱NF-κB,IL-6,IL-1β的表达,降低结肠黏膜LPO含量,增加红细胞SOD含量,进而增加机体对O2-的清除,从而达到促进大便乳酸排泄,减少结肠炎症损伤有关.%Objective; To study the mechanism the Meizhen Granule on the ulcerative colitis. Methods: Models established by the DNCB were used to detect the colon mucosa histopathology.the colon injury score,the SOD and ihe LPO in the colonic mucosa. Results: In the treatment group,the changes of the colon mucosa histopathology were obvious compared with the control group. Meanwhile,the colon injury score,the stool lactic acid level and the LPO concentration were decreased significantly. However,the SOD level in the treatment group was increase obviously. Some indices such as NF-KB,IL-6 and IL-lβ were reduced obviously in the Meizhen Granule group. There were some statistical differences in above indicators between the Meizhen Granule group and the SASP group. Conclusion: The mechanism of Meizhen Granule against the ulcerative colitis could be related that it can reduce expressions such as NF-KB,IL-6,IL-1Β and LPO. It can increase the SOD level and O2~ clearance to improve the stool lactic acid excretion and decrease the colon inflammatory injury.
