不同月龄阶段陕北白绒山羊生精细胞发育规律研究

         

摘要

In order to explore the spermatogenic cell development of Shaanbei white cashmere goats at different developmental stages,the testis of Shaanbei white cashmere goats (one-,three-,six-month-old and adult goats) were collected and cut into pieces for staining using hematoxylin-eosin.The results showed that (1) The diameter of goat seminiferous tubules increased significantly with age (P<0.05).The difference in diameter between 3-month-old and adult goats was not significant,representing 150.72 μm and 161.94 μm,respectively(P>0.05).(2)The cell layer of goats also increased with age,the cell layer of 6-month-old goat (7.70 layers) was significantly higher than that of one-and three-month-old goats,respectively (P<0.05).The total number of cross-sectional cells in seminiferous tubules both of six-month-old and adult goats was strikingly different compared to those of 1-month-old and 3-month-old cashmere goats (P < 0.05).(3) Moreover,the proportion of spermatogonia of 3-month-old goats (46.40%) was significantly higher than that of one-,six-month-old and adult goats,just about 33.10%,9.10% and 7.60%,respectively (P<0.05).(4)The expression of PLZF in testis decreased significantly with age (P<0.05).Marker genes of SSCs,such as CD49f,PLZF,Thy1,ETV5,VASA,Oct4,Nanog and Nanos,were highly expressed in the testis of 3-month-old goats,and except for the CD49f gene,expression levels of other genes were significantly higher than those of other groups (P<0.05).The results showed that the cashmere goats reached sexual maturity in 6 month old,and the most abundant spermatogonia were in 3-month-old testis.%为研究陕北白绒山羊生精细胞发育规律,取1月龄、3月龄、6月龄及成年陕北白绒山羊睾丸组织,HE染色观察睾丸发育情况.结果表明:(1)随着年龄的增长,曲细精管直径显著增大(P<0.05),6月龄达到150.72 μm,略低于成年羊(161.94 μm) (P>0.05).(2)细胞层数随年龄的增长而增长,6月龄达到7.70层,显著高于1月龄和3月龄(P<0.05).曲细精管横截面细胞总数随年龄增长而增长,6月龄和成年绒山羊曲细精管横截面细胞总数显著高于1月龄和3月龄绒山羊(P<0.05).(3)3月龄陕北白绒山羊曲细精管中精原细胞最为丰富,占总细胞数的46.40%,明显高于1月龄(33.10%)、6月龄(9.10%)和成年羊(7.60%)(P<0.05).(4)精原干细胞标记基因(CD49f、PLZF、Thyl、ETV5、VASA、Oct4、Nanog、Nanos2)均在3月龄睾丸中高表达,其中Oct4、PLZF、Thy1、ETV5、VASA、Nanog及Nanos2显著高于1月龄、6月龄及成年羊(P<0.05).研究结果表明,6月龄陕北白绒山羊已经达到性成熟;与1月龄、6月龄和成年羊相比,3月龄陕北白绒山羊睾丸组织含有更加丰富的精原细胞.
