MTDH、Tmem16a在脉络膜黑色素瘤中的表达及意义

摘要

目的 研究异粘蛋白(MTDH)和跨膜蛋白16a (Tmem16a)在脉络膜黑色素瘤中的表达及临床意义.方法 选择25例(25只眼)确诊的脉络膜黑色素瘤术后组织为实验组,另取瘤旁正常脉络膜组织15例(15只眼)作为对照组,采用免疫组化法检测其MTDH 、Tmem16a蛋白表达情况,分析MTDH、Tmem16a蛋白表达情况和患者临床特征之间的关系.结果 实验组MTDH、Tmem16a阳性表达率分别为60％、52％,均明显高于对照组(P＜0.05).MTDH、Tmem16a蛋白表达与脉络膜恶性黑色素瘤巩膜浸润程度及远处转移相关(P＜0.05);与年龄、性别、肿瘤大小无关(P＞0.05).结论 MTDH、Tmem16a在脉络膜黑色素瘤中呈高表达,且与巩膜浸润程度及远处转移相关,可能成为临床诊治脉络膜黑色素瘤的重要指标.%Objective To investigate the expression and clinical significance of MTDH and transmembrane protein 16A (Tmem16a) in choroidal melanoma.Methods Postoperative tissuesfrom 25 cases of choroidal melanoma were collected (experimental group),and adjacent normal choroidal tissues in 15 cases were collected as control.MTDH and Tmem16a protein expression was tested,and the relationship between MTDH and Tmeml6a were analyzed.Results Positive expression rates of MTDH and Tmem16a in the experimental group were 60％ and 52％,respectively,which were significantly higher than those in the control group (P ＜ 0.05).The expression of MTDH and Tmem16a were related to the degree of scleral invasion and distant metastasis in patients with choroidal malignant melanoma (P ＜ 0.05).The relationship was independent of age,sex and tumor size (P ＞ 0.05).Conclusions MTDH and Tmem16a are highly expressed in choroidal melanoma and related to the degree of invasion and distant metastasis of the sclera,which may become an important indicator for the diagnosis and treatment of choroidal melanoma.
