津力达颗粒对2型糖尿病大鼠骨骼肌氧化应激水平以及 SIRT3表达的影响

摘要

Objective To test the effects of Jinlida Granules on the muscle oxidative stress and the expression of SIRT3 in diabetic rats. Methods 30 SD rats were randomly divided into control group,model group and Jinlida Granules groups. Rats in the control group were treated with normal diet. Model group were treated with high sugar and high fat diet and streptozotocin. The sugar level and insulin level were measured to assess the success of model. Jinlida Granules groups were treated with different dose of Jinlida Granules[0. 75 g/(kg·d),1. 5 g/(kg·d) and 3. 0 g/(kg·d)]. Eight weeks later,the expression of SQD,GSH,RQS,MDA were tested by ELISA kit. The mRNA and protein expression of SIRT3 were measured by PCR and Western - blot respectively. Results GSH,SQD and SIRT3 activity in model group were significantly de-creased,and MDA and RQS level increased. After Jinlida Granules treatment,GSH and SQD activity as well as SIRT3 were increased,MDA and RQS level were reduced( P < 0. 05). Conclusion Jinlida Granules can regulate the oxidative stress and up regulate the expression of SIRT3 to modulate the level of blood glucose and insulin resistance.%目的：探讨津力达颗粒对2型糖尿病大鼠骨骼肌氧化应激水平以及乙酰化酶3（SIRT3）表达的影响。方法50只 SD 大鼠随机分为对照组、模型组以及津力达颗粒组，每组10只。对照组大鼠给予普通饲料，模型组喂以高糖高脂饲料并给予链脲佐菌素并通过血糖水平以及胰岛素水平验证模型。津力达颗粒组分别给予0.75 g/（kg·d）、1.5 g/（kg·d）以及3.0 g/（kg·d）三种不同剂量津力达颗粒。给药8周后测定各组血糖水平以及空腹胰岛素水平，大鼠肌肉中超氧化物歧化酶（SQD），谷胱甘肽过氧化物酶（GSH），活性氧（RQS）活性及丙二醛（MDA）含量；PCR 以及 Western- blot 法检测 SIRT3蛋白表达。结果与对照组比较，模型组 GSH、SQD 以及 SIRT3活性显著降低，而 MDA 以及 RQS含量升高；津力达颗粒治疗后，GSH、SQD 活性以及 SIRT3升高，MDA 以及 RQS 含量降低（ P ＜0.05），津力达颗粒可以降低模型组血糖以及空腹胰岛素水平，其中高剂量的效果要优于低、中剂量。结论津力达可通过抗氧化应激以及上调 SIRT3降低血糖水平以及胰岛素抵抗，且其效果呈剂量依赖性。
