碳酸镧诱导人骨骼肌细胞胰岛素抵抗模型构建

             

摘要

目的 探讨碳酸镧处理人骨骼肌细胞能够否建立骨骼肌细胞胰岛素抵抗模型.方法 将配制好的成人骨骼肌细胞细胞直接在培养基中进行培养,培养基样板为96孔.将细胞分为对照组、胰岛素组和碳酸镧组,后两组分别添加不同浓度的胰岛素(5 ×10-7μmol/L、1×10-7μmol/L)和碳酸镧(5μmol/L、10μmol/L、100μmol/L、1 000 μmoL/L).培养6h、12h、24 h、36 h后,使用葡萄糖氧化酶法测定葡萄糖浓度.结果 培养24 h后,10 μmol/L、100 μmol/L、1 000 μmol/L碳酸镧组及5×10-7 μmol/L胰岛素组中,葡萄糖含量与对照组细胞相比均有不同程度的增加(P<0.05).在100 μmol/L、1 000 μmol/L的碳酸镧培养基中培养的时间达到6h后,胰岛素将会对溶液中葡萄糖的浓度产生较大的影响.结论 碳酸镧处理骨骼肌细胞能够引发胰岛素抵抗,可为研究转胰岛素抵抗发病机制和药物筛选提供新的骨骼肌细胞胰岛素抵抗模型.%Objective To investigate the possibility of lanthanum carbonate on induction of insulin resistance model in skeletal muscle cells.Methods The adult skeletal muscle cell cells were inoculated into a 96-well plate.Cells were divided into blank group,insulin group and lanthanum carbonate group.Cells in insulin and lanthanum carbonate groups were treated by different doses of insulin (5 × 10-7 μmol/L and 1 × 10-7 μmol/L) and lanthanum carbonate (5 μ mol/L,10 μ mol/L,100 μ mol/L,1 000 μ mol/L).The glucose concentration was measured at 6 h,12 h,24 h and 36 h,respectively.Glucose oxidase method was used to determine glucose content.Results Our study showed that,after 24 h,10 μmol/L,100 μmol/L,1 000 μ mol/L lanthanum carbonate group and 5 × 10-7 μmol/L insulin glucose contents in culture medium were higher than normal group and high cell difference (P <0.05).Conclusion Lanthanum carbonate can induce insulin resistance model in skeletal muscle ceils,providing a novel cellular model for study of pathogenesis of insulin resistance and development of new drags.
