比较高碘酸-雪夫与双花扁豆凝集素反应标记的小鼠蜕膜自然杀伤细胞

         

摘要

目的 对比高碘酸-雪夫(Periodic acid-Schiff,PAS)与双花扁豆凝集素(Dolichos Bifows Agglutinin,DBA)组织化学法所显示的小鼠蜕膜自然杀伤 (decidual natural killer,dNK) 细胞的不同.方法 取妊娠第3~19天小鼠子宫,制备成连续的切片,分别经PAS、DBA组织化学法、DBA加PAS双重反应显示dNK细胞.结果 典型的 dNK细胞含有PAS阳性颗粒,其胞膜、核膜和颗粒膜与DBA特异性结合而呈阳性反应;妊娠早期(妊娠第3~6天)不含颗粒的幼稚dNK细胞也可以被DBA标记.DBA加PAS双重反应时,含颗粒的dNK细胞大部分可以被同时标记.结论 DBA是dNK细胞的一种新的标志物,在妊娠早期可用以显示 PAS阳性颗粒尚未出现的幼稚dNK细胞.%To identify the difference of mouse decidual nalural killer (dNK) cells reacting to Periodic Acid Schiffs (PAS) and Dolichos Biflorus agglulinin (DBA) . Methods The dNK cells of mice uleri Gd 3 lo Gd 19 (using the copulalion plug as Gdl) were shown on conlinuous paraffin seclions by PAS, DBA, PAS/DBA double reaclions prolocol. Results Typical dNK cells conlained numerous PAS posilive reaclive cyloplasmic granules. The cyloplasmic membrane, nuclear membrane and granule membrane of dNK were posilive reaclion lo DBA due lo their specific binding. The immature dNK cells in the ulerus of early pregnancy (on Gd 3 lo Gd 6) were negative lo PAS because of iheir absence of granules. Majority of dNK cells could be labeled at the same lime with DBA plus PAS double staining. Conclusion DBA can be used as a new marker lo show dNK due to its specific binding to membranes. DBA has advantage especially in early pregnancy (such as on particles Gd 3) because PAS posilive granules have not yet appeared in immature dNK cells.
