大黄鱼Follistatin基因克隆及表达分析

         

摘要

Follistatin (FST), belonging to transforming growth factor (3 superfamily, plays an important role in animal muscle growth. RT-PCR, RACE and conventional PCR techniques were used to clone FST gene from Larimichthys crocea in this study. The obtained FST gene was 3195 bp in length containing a 96 bp 5' untranslated region(UTR), five exons, four introns, and a 47 bp 3'-UTR. The complete open reading frame of FST was 972 bp, encoding a peptide of 323 amino acid residues. It was consisted of signal peptide of 31 amino acid residues and mature peptide of 292 amino acid residues. FST is highly conserved in the evolution, and the nucleotide and protein sequence similarity between Larimichlhys crocea and Sparus aurata reached 96% and 99%, respectively. FST gene was expressed in various tissues of Larimichthys crocea, in which the gill tissue is highest, and the spleen the lowest. When water temperature changed, FST expression in eye and muscle tissues greatly changed. It suggested that FST may play an important role in fish growth and development.%卵泡抑素(follistatin,FST)是转化生长因子β(Transforming growth factor-β)超家族成员之一,在动物肌肉生长中起重要作用.采用RT-PCR、RACE和常规PCR技术克隆了大黄鱼FST基因.获得的基因序列长3195 bp,其中5'非编码区96 bp,3'非编码区47 bp,含5个外显子及4个内含子.该基因开放阅读框972 bp,编码323个氨基酸,其中信号肽31个氨基酸,成熟肽292个氨基酸.Blast结果表明,大黄鱼FST基因与金鲷FST基因的核苷酸及蛋白序列同源性最高,分别达到96%和99%.FST基因在大黄鱼脑、眼、鳃、肾等多个组织中表达,其中鳃组织的表达量最高,脾组织中表达最低.检测水温19℃、25℃、30℃时大黄鱼不同组织FST基因表达量,眼和肌肉组织中FST基因表达量变化显著,推测FST基因可能在鱼类生长发育中发挥重要作用.
