DNA2基因表达与乳腺癌恶性程度及雌激素受体表达的关系

         

摘要

Objective:To explore the role of DAN2 expression in breast cancer maliganant degree and positive estrogen receptor ( ER) . Methods:The DNA2 mRNA in carcinoma tissue and para-carcinoma tissue of 132 breast cancer patients were extracted,and the expression level of DNA2 mRNA was detected using real-time quantitative PCR. The relationships between ER,and DNA2 expression and degree of malignancy were analyzed. The expressions of DNA2 protein in ER positive and ER negative cells were detected,which was used to verify the positive correlation between DNA2 and ER. Results:The positive ER was positive correlation with DNA2 expression,and the positive ER could up-regulate the DNA2 expression. The positive ER was not correlation with tumor malignance,and the positive ER upregulated the DNA2 expression to increase the MCF7 cells resistance to CPT. Conclusions:The regulation of the DNA2 expression can significantly promote the metastasis of malignant tumor,and the positive ER up-regulates the DNA2 expression to increase the drug resistance of tumor cells.%目的:探究DNA2基因表达水平与乳腺癌恶性程度及雌激素受体表达的关系.方法:提取132例乳腺癌病人肿瘤组织和癌旁组织中DNA2的mRNA,通过实时定量PCR检测DNA2表达水平的变化.通过统计病人ER是否阳性,以及乳腺癌细胞恶性程度等信息,结合DNA2表达量差异,分析ER对DNA2表达的调控关系以及这种调控与肿瘤恶性程度的关系.并通过培养ER阳性和阴性的细胞来检测DNA2蛋白表达量,以及分析细胞存活来验证相关性分析结果.结果:DNA2高表达与恶性程度高度正相关.ER阳性与DNA2高表达正相关,ER阳性可以上调DNA2表达量.ER阳性与肿瘤高恶性程度不相关,而ER阳性能通过上调DNA2表达量显著增加MCF7细胞对喜树碱的耐药性.结论:DNA2调控对肿瘤恶性转移有显著的促进作用,ER阳性上调DNA2表达量增加肿瘤细胞耐药性
