菊花茎尖克隆体外繁殖研究

     

摘要

[目的]研究不同因子对菊花芽苗体外繁殖的影响.[方法]以秋菊品种“绣球”和“妃子笑”的项茅或丰木质化茎段为外植体,研究不同激素及组合对菊花芽苗体外繁殖的影响.[结果]在MS+ BA 0.5 mg/L+ IBA 0.1 ng/L的诱导培养基上两品种均能诱导分化出芽.在芽的生长增殖阶段,接种到MS+BA 1.5 mg/L+IBA 0.5 mg/L+GA0.1mg/L的增殖培养基上,“红绣球”和“妃子笑”的增殖系数分别可达6.8和6.4,芽苗生长速度快,生长健壮.在1/2 MS+ IBA 0.5 mg/L的生根培养基上,生根所用时间短,根系数量大且粗细始终,移栽成活率较高.[结论]该研究为“绣球”和“妃子笑”菊花品种的组培快繁提供了理论与技术支持.%[Objective]The research aimed to study the influences of different factors on the in vitro propagation of shoots of Dendranthema morifolium. [ Method] By using the apical buds and cutting stems with buds of two chrysanthemum species of " Xiuqiu" and " Feizixiao" as ex-plants, the effects of different hormones recombination at different concentration on the clone propagation in vitro were studied. [ Result]The multiplication rate of adventitious buds from explants of " Xiuqiu" and " Feizixiao" can reach 6.8 and 6.4 respectively on the mediums supplemented with BA 1.5 mg/L, IBA 0.5 mg/L and CA 0. 1 mg/L. Furthermore, the condition of chrysanthemum shoots root induction had been studied. The results indicated that the best condition of rooting is 1/2MS basal medium supplemented with 0.5 mg/L IBA. [Conclusion]This research could provide scientific and technological basis for the tissue culture and rapid propagation of "Xiuqiu" and "Fei zixiao".
