铬胁迫对小麦幼苗叶片DNA含量的影响

     

摘要

[目的]研究Cr(6+)对小麦幼苗叶片DNA含量的影响.[方法]用5~100 mg/L Cr(6+)分别处理3和10d龄小麦幼苗,紫外分光光度法和电泳法测定DNA含量变化.[结果]Cr(6+)处理的小麦幼苗叶片DNA含量显著降低,3d龄幼苗叶片DNA含量的下降幅度大于10 d龄幼苗.电泳图谱表明,对照叶片DNA带基本完整,而Cr(6+)处理的DNA则发生了降解,出现拖尾现象.[结论]Cr(6+)处理减少了小麦幼苗叶片DNA含量,进而影响其正常生长.%[Objective] To study the effect of Cr6+ on the DNA content in leaves of wheat seedlings. [ Method] 3-day-old and 10-day-old wheat seedlings were treated separately with 5-100 mg/L Cr6+ , after that the DNA content was determined by UV spectrophotometry method and agarose gel electrophoresis. [Result] The content of DNA in leaves of Cr6+ treated wheat seedlings all reduced significantly, the decrease of DNA content in 3-day-old seedlings was bigger than that in 10-day-old seedlings. Agarose gel electrophoresis indicated that DNA belt was basic complete in leaf of control seedling, but the DNA belts in leaf of Cr6+ treated seedling had the degradation with the trailing phenomenon. [Conclusion] Cr6+ treatment led to the decrease of DNA content in leaves and affected the abnormal growth of wheat seedlings.
