五加皮多糖提取工艺的优化及其性能分析

     

摘要

[ Objective ] To optimize the extraction conditions of CORTEX ACANTHOPANACIS polysaccharide and analyze its property, so as to provide basis for the development and application of CORTEX ACANTHOPANACIS polysaccharide. [ Method] By employing single factor test and orthogonal test, the extraction process of CORTEX ACANTHOPANACIS polysaccharide was optimized, and purified with concentration, dialysis and ethanol precipitation, the solubility, reducing sugar and protein of CORTEX ACANTHOPANACIS polysaccharide were de tected, in the end, the CORTEX ACANTHOPANACIS polysaccharide was identified by IR spectrum. [ Result] The optimum extraction conditions of CORTEX ACANTHOPANACIS polysaccharide were: 90 ℃ of extraction temperature, three times of extraction times, solvent quantity of 100 and extraction duration of 2.0 h. Yield of CORTEX ACANTHOPANACIS polysaccharide was up to 9.38 % under the optimal extraction process. Good in solubility, rich in starch and low in reducing sugar, the CORTEX ACANTHOPANACIS polysaccharide was typical polysaccharide. [ Conclusion] The optimal extraction conditions for CORTEX ACANTHOPANACIS polysaccharide and its properties are obtained in this study, which provides basis for the development and application of CORTEX ACANTHOPANACIS polysaccharide.%[目的]优化五加皮(CORTEX ACANTHOPANACIS)多糖的提取工艺,并分析其性能,为五加皮多糖的开发利用奠定基础.[方法]用单因素试验和正交试验优化五加皮多糖的提取工艺;采用浓缩、透析和醇沉等对多糖进行纯化,对其溶解性、还原糖含量、蛋白质进行检测,并进行红外鉴定.[结果]五加皮多糖提取的最佳工艺为:温度为90℃,提取次数为3次,溶媒量为100,提取时间为2.0 h.在最佳提取工艺下,五加皮多糖的提取率为9.38%.该多糖溶解性较好,含有淀粉,含有少量还原糖,具有典型的多糖特征光谱.[结论]该研究优化了五加皮多糖的提取工艺,并对其性能进行了分析,为五加皮多糖的开发利用奠定了基础.
