天津海底淤泥微生物DNA的提取方法研究

         

摘要

[目的]探索一种快速、高效提取海底淤泥中微生物DNA的最佳方法,为开展海底淤泥微生物的分子生物学研究奠定基础.[方法]分析了不同DNA提取液浓度和不同的腐植酸去除方法对淤泥中微生物DNA提取的产率、纯度的影响,探讨天津市海底淤泥中微生物DNA提取的最佳方法.[结果]海底淤泥用焦磷酸钠和氢氧化钠混合浸提剂提取腐植酸后,再用蒸馏水洗去浸提液,用4ml DNA提取液提取,所得DNA的产率及纯度均优于对照组.[结论]该试验结果为海底淤泥微生物的分子生物学研究奠定了基础.%[Objective] To explore a rapid and high-efficient method to extract microbial DNA from the seabed sludge of Tianjin, so as to lay foundation for studying the molecular biology of microorganism in the sludge. [ Method] The influence of different DNA extracting concentration and different humic acid removal methods on the yield and purity of microbial DNA from sludge was analyzed to explore the best extraction method. [ Result] The humic acid was extracted from seabed sludge with the mixed sodium pyrophosphate and sodium hydroxide, and then the extracting solution was washed by distilled water, and then extracted with 4 ml of DNA extracts, the yield and purity of DNA were better than the control plot. [ Conclusion ] The experimental results lay a foundation for studying the molecular biology of microorganism in the seabed sludge.
