胆总管结扎和四氯化碳诱导小鼠肝纤维化模型的研究

         

摘要

目的:比较胆总管结扎(BDL)与 CCl4诱导小鼠肝纤维化的生化指标、肝脏病理学指标及α-Sma 的表达。方法:60只健康雄性 BalB/C 小鼠,分为 CCl4肝纤维化模型组(20只)及其对照组(10只)和 BDL 肝纤维化模型组(20只)及其对照组(10只)。观察小鼠一般情况,生化方法测血清 ALT、TBA 水平。HE 和 Masson 染色观察肝组织的病理变化及免疫组织化学染色观察肝组织α-Sma 的表达。结果:血清生化指标显示两组模型血清的 ALT 酶活性都明显高于对照组,肝组织的病理指标 Masson 染色及免疫组化检测均显示模型组的蓝色染色坏死区域明显多于对照组。结论:两种处理方法均可诱导小鼠肝纤维化,且 BDL 较早引起肝功能及肝纤维化指标升高。%Objective To compare serum biochemical parameters,liver pathology,and α-Sma expression in BalB/C rats with hepatis fibrosis induced by bile duct ligation(BDL)and carbon tetrachloride(CCl4 ).Method Sixty healthy male BalB/C rats were devided into CCl4 model(n =20),CCl4 control(n =1 0),BDL model(n =20),BDL control(n =1 0),General condi-tions of rats were examined,ALT,TBA were measured by biochemical analysis.Pathological changes in liver tissue were exam-ined through hematoxylin eosin and Masson staining.Expression of α-Sma was assayed by immunohistochemistry.Re-sults Serum biochemical analysis showed that ALT and TBA levels in two model groups were obvious higher than those control groups,Pathological changes in liver tissue(Masson staining and immunohistochemistry of α-Sma)also showed that the blue staining of two model groups were significantly higher than those control groups.Conclusion Both BDL and CCl4 can induce he-patic in rats,The former induces early increase in the indicators of liver function and hepatic fibrosis.
