豚鼠肝细胞大鼠脾内移植治疗暴发型肝衰的实验研究

摘要

目的探讨异种肝细胞移植治疗暴发型肝衰的疗效。方法杂种豚鼠为供体，胶原酶消化法制备肝细胞。SD大鼠为受体，D-氨基半乳糖（D-GI）腹腔内一次注射制作肝衰模型。48小时后将豚鼠肝细胞（1.5×107个）移植于大鼠脾内。同种移植及生理盐水为对照。移植后观察受体二周存活率，在不同时间作移植物病理及组织化学检查。结果受体2周存活率：异种移植组71％，同种组68％（P＞0.0 5），生理盐水对照组25％（P＜0.01）。豚鼠肝细胞移植后12～24小时其结构和功能基本保存完好。结论与同种移植一样，异种肝细胞移植能逆转大鼠暴发型肝衰%Objetive　To observe the e ffic acy of xenogenic hepatocyte transplantai on（XHX） in a toxic model of fulminant he patic failue（FHF） in rat．Method　 SD rats（n＝14） that received 1.5×107 g uinea pig hepatocyte intraspleen at 48ho urs after D-galactosamine（D-GI） intraper itnear injection （1.95mg／kg）．The two week survival rate was compared with s ynge nic hepatocy te transplantation（n＝16）and control an imals（n＝16）．The pathological examinati ons were done at the different time afte r transplantion．Results　The XHT was able to significantly improve surviv al rate as S HT done（71％versus 68％，P＞0.05）when comp ar ed with functions of guinea pig hepatocy tes transplanted were basiclly normal du ring the time of 12-24 hours after trans plantation．Conclusion　XHT could reverse the FHF of rat induced by D-GI
