地塞米松对人羊膜上皮WISH细胞糖皮质激素受体α表达的影响

     

摘要

Objective To observe the effects of dexamethasone on glucocorticoid receptor-a (GR-a) expression in the human amniotic epithelial cells, WISH cells. Methods The WISH cells were cultured in vitro with different concentrations of dexamethasone( 10~8,10~7,10~6,10~5,10~4 mol/L) for 48 h (group A) and a single concentration of dexamethasoneQCT6mol/L) for different times of 24,48,and 72 h respectively. The results were compared to the blank control groups. The GR-a mRNA expressions in WISH cells were detected with RT-PCR. Results The expression of GR-a mRNA of WISH cells treated with 10~4mol/L DEX was higher than that of control group, which was loweer in those treated with the other concentration groups. The expression of GR-a mRNA was siginificantly lower in the cells treated with different times than that in those of blank controls (P<0. 01). Conclusion The expression of GR-a mRNA is decreased when treated with low concentration of dexamethason(5^ 10~6 mol/L), which is increased when treated with high concentration of it (≥10~5mol/L). The decrease of GR-a mRNA expression is not reduced as the treating time prolongs. The clinical, dose of dexamethasone for promoting lung paturity of the fetus could not lead to the onset of delivery.%目的 观察地塞米松对体外培养人羊膜上皮WISH细胞糖皮质激素受体(GR)-α表达的调节.方法 将体外培养的WISH细胞分为不同浓度地塞米松(10-8、10-7、10-6、10-5、10-4 mol/L)作用48 h组,和单一浓度地塞米松作用不同时间组(终浓度10-6mol/L,培养24、48、72 h).两组均设空白对照组.采用RT-PCR方法检测各组细胞中GR-a mRNA的表达.结果 在不同浓度地塞米松作用48 h后,10-4mol/L组细胞GRα mRNA表达强于空白组,其余各组细胞GR-α mRNA表达均弱于与空白组(P<0.01).在地塞米松作用不同时间组,各亚组细胞GR-α mRNA表达与空白组相比,均明显减弱(P<0.01).结论 低浓度地塞米松可下调WISH细胞中GR-α mRNA表达;高浓度地塞米松可上调GRα mRNA的表达.GR-α mRNA表达不随地塞米松作用时间的延长而下降.临床上常规剂量和疗程应用地塞米松促胎肺成熟时,不会导致促分娩发动作用.
