重组MMP-2对人肾系膜细胞Fkn基因表达和分泌的影响

Objective To observe the effects of matrix metalloproteinase-2(MMP-2) on the expression of fractalkine(Fkn) in cultured human renal mesangial cells(HRMCs). Methods HRMCs were incubated with different concentrations of MMP-2. Fkn mRNA expression of HRMCs was analyzed by RT-PCR and the concentration of Fkn in the supernatant was quantified by ELISA.Results Fkn mRNA expression of HRMCs increaed as MMP-2 increased (P＜0. 05). The expression of Fkn protein in the supernatant decreased as MMP-2 increased (P＜ 0. 05). Conclusion MMP-2 could increase the Fkn mRNA expression in HRMCs, but decrease the concentration of Fkn in the supernatant.%目的 观察重组基质金属蛋白酶-2(MMP-2)对人肾系膜细胞(HRMC)趋化因子FknmRNA表达的影响.方法 用重组MMP-2干预体外培养的HRMC,逆转录PCR检测Fkn mRNA表达,ELISA法检测HRMC上清液中Fkn蛋白含量.结果 HRMC的Fkn mRNA表达水平随MMP-2浓度增加而增加(P<0.05),上清液中Fkn蛋白含量随MMP-2浓度增加而降低(P<0.05).结论 重组MMP-2可以增加Fkn mRNA的表达,而降低上清液中Fkn蛋白含量.
