α1，3-岩藻糖基转移酶-Ⅴ在类风湿性关节炎滑膜组织中的表达及意义

摘要

目的探讨α1，3‐岩藻糖基转移酶‐Ⅴ（FucT‐Ⅴ）在类风湿性关节炎（RA ）患者膝关节滑膜组织中表达及意义。方法取17例RA患者（实验组）和9例半月板损伤行关节探查手术患者（对照组）的滑膜组织，用苏木素伊红染色观察滑膜组织的特征及炎症细胞浸润情况，Western blot检测FucT‐Ⅴ蛋白在滑膜组织中的表达，免疫组化检测FucT‐Ⅴ在滑膜组织中的表达及分布情况，免疫荧光法观察检测 FucT‐Ⅴ与 RA滑膜主要炎症细胞共定位情况。结果实验组患者滑膜组织中FucT‐Ⅴ蛋白的表达高于对照组（P＜0．05）。在RA滑膜炎症过程中，FucT‐Ⅴ与滑膜组织中巨噬细胞、成纤维细胞、中性粒细胞和活性T淋巴细胞存在共定位。结论 FucT‐Ⅴ可能参与调节RA滑膜组织的炎症反应过程。%Objective To investigate the expression and significance ofα1 ,3‐fucosyltransferase‐V(FucT‐Ⅴ) in knee synovial tissue in the patients with rheumatoid arthritis(RA) .Methods Synovial tissue samples were obtained from 17 RA patients(group R) and 9 patients(group C) undergoing arthroscopy surgery for meniscus crush .The chacacteristics and inflammatory cell infiltration of synovial tissues were observed by hematoxylin‐eosin staining ,the expression of FucT‐Ⅴ protein was detected by Western blot ,and cellular colocalization of FucT‐Ⅴ and inflammatory corpuscle were analyzed by double immunofluorescence and immunohistochemical .Results The expression of FucT‐Ⅴwas higher in group A than that in group C(P<0 .05) .In group A ,FucT‐Ⅴ was mainly expressed in macrophages ,fibroblasts ,neutrophils ,and activated T lymphocytes .Conclusion FucT‐Ⅴ may play an important role in the inflammatory processes in synovial tissue of RA patients .
