全反式视黄酸及干扰素对胃癌细胞系MKN45中p16、p21、c-myc基因表达的影响

         

摘要

Objective To explore the influence of all-trans-retinoic acid and interferon on Gastric Carcinoma MKN45 cells. Methods Retinoic acid and interferon were added into the Cell Line of Gastric Carcinoma MKN45 at the same time,after cell culture,MTT were perfomed to determinate the cell growth conditions and detect the expression of p16,p21 and c-myc by Northern blot and immunohistochemical methods. Results Interferon (IFN) coordination of all-trans retinoic acid (ATRA) could effectively inhibit the growth of MKN45 cells,cancer cells by the combination therapy induced the expression of p16 and p21 genes to raise the level, c-myc gene expression decreased. RARα retinoic acid receptor gene in MKN45 cells showed low expression of IFN induced by ATRA and fhe expression level of increase. Conclusion ATRA combined IFN-induced p16 and p21 can regulate gene expression,inhibit the growth of gastric cancer MKN45 cells,which may be related to a high level of expression of cell RARα gene.%目的 探讨全反式视黄酸(ATRA)及干扰素(IFN)对胃癌MKN45细胞的影响.方法 将ATRA及IFN同时加入胃癌细胞系MKN45培养体系中,MTT法测定细胞生长状况,Northern Blot和免疫组化测定p16、p21及c-myc表达情况.结果 IFN、ATRA联合用药可有效抑制MKN45细胞生长,细胞p16和p21表达水平升高,c-myc表达水平下降.视黄酸受体RARα在MKN45细胞中呈低水平表达,经ATRA和IFN诱导后表达水平升高.结论 ATRA、IFN联合用药可调节p16和p21表达水平,抑制胃癌MKN45细胞生长,可能与RARα高水平表达有关.
