白细胞介素-6对兔血管平滑肌细胞产生C反应蛋白的影响

         

摘要

Objective:To investigate whether interiukin-6 (IL-6) can induce the primarily cultured rabbit vascular smooth muscle cells (VSMCs) to produce C-reactive protein (CRP). Methods: Rabbit primary VSMCs from abdominal aorta were cultured in vitro. Then IL-6 (10ng/ml), internukin-1β (IL-1β, 25ng/ml), IL-6 (10ng/ml) + IL-1β (25ng/ml) were used respectively to stimulate the primary cells for 48h. The VSMCs were incubated with the solvent of IL-6 in the control group. Reverse-transcriptase polymerase chain reaction (RT-PCR) and western blot were used to detect CRP mRNA and protein expression of the primary cells respectively. Results: Incubating with IL-6 alone after 48h, CRP mRNA level was (3. 50 ± 1. 17) fold of the control and CRP was modestly produced. The difference was significant comparing with the control. Stimulating with the combination of IL-6 and IL-1β, CRP mRNA level was (5.92 ± 1. 30) fold of the control and CRP was maximally produced, whereas incubation with IL-1β alone did not induce CRP production. Conclusion: IL-6 can induce the primary rabbit VSMCs to produce CRP. IL-1β can significantly enhance the induction of CRP by IL-6.%目的:探讨白细胞介素-6(IL-6)能否诱导体外原代培养的兔血管平滑肌细胞(VSMCs)产生C反应蛋白(CRP). 方法:体外原代培养新西兰兔腹主动脉VSMCs.分别用白细胞介素(IL)-6( 10 ng/ml)、IL-1β 25 ng/ml或IL-6(10 ng/ml)+IL-1β(25 ng/ml)刺激VSMCs 48 h.对照组用等体积的IL-6的溶剂孵育48 h.反转录聚合酶链反应(RT-PCR)和免疫印迹法(Western blot)分别检测培养细胞CRP mRNA和蛋白的表达情况. 结果:对照组的原代VSMCs未见CRP的产生.VSMCs与IL-6单独孵育48 h后,CRP mRNA的表达量是对照组的(3.50±1.17)倍,能产生少量CRP,与对照组相比有差异.单独应用IL-1β不能诱导VSMCs产生CRP,但是在IL-1β和IL-6的联合刺激下,CRP mRNA表达量是对照组的(5.92±1.30)倍,CRP的合成亦达最大值. 结论:IL-6能诱导原代培养的兔VSMCs产生CRP.尽管单独应用IL-1β不能刺激VSMCs产生CRP,但是它却能显著增强IL-6诱导CRP的表达.
