HSP70在STZ-糖性白内障发病机制中的研究

     

摘要

·AIM: To evaluate the role of heat shock protein (HSP70) in the p athogenesis of streptozotocin (STZ)-diabetic cataract. · METHODS: A total of 66 SD rats were randomly divided into two groups: Normal Control Group and Cataract Group. Diabetic cataract was induced by STZ intraperitoneal injection (70mg/kg in 0.1 mol/L sodium citrate buffer). Lenses were examined once a week with a slit lamp. Eyeballs were isolated at the end of 2, 4 and 8weeks after STZ injection. Immunohistochemical staining was used to detect the expression of HSP70 in both normal and diabetic LECs.· RESULTS: The lenses in Control Group maintained completely transparent throughout the experimental period. Those in Cataract Group began to exhibit a vacuolar stage by 2wk and near opacity by 8wk. The expression of HSP70 was significantly different between normal and diabetic LECs, and was correlated with the development of cataract.· CONCLUSION: HSP70 may play a critical role in the pathogenesis f diabetic cataract by modulating the growth of LECs.%目的:研究HSP70在链脲佐菌素-糖性白内障发生发展中的作用.方法:将66只SD大鼠随机分为2组,正常对照组与白内障组.用链脲佐菌素(STZ)诱发糖性白内障,每周观察晶状体的变化,在实验开始后2,4,8周末,分别摘取眼球,采用免疫组化技术检测热休克蛋白-70(HSP70)在晶状体上皮细胞(LECs)中的表达.结果:对照组晶状体一直保持透明,白内障组晶状体在2周末出现空泡,8周末全部混浊.HSP70在对照组中未见表达,在白内障组中表达明显,并随着白内障的发展而表达增加.结论:HSP70可能通过调节晶状体上皮细胞的生长在糖性白内障的发生、发展中起重要作用.
