活血熄风方对MCAO局灶性脑缺血模型大鼠脑组织Caspase-3蛋白表达的影响

摘要

目的:观察活血熄风方对局灶性脑缺血大鼠的神经保护作用,并进一步探讨其作用机制.方法:采用线栓法阻断大鼠一侧大脑中动脉(MCAO),制备局灶性脑缺血模型,观察活血熄风方对MCAO模型大鼠脑缺血后神经功能缺损导致的行为学变化,并以免疫组化法检测对其大脑皮质细胞凋亡相关蛋白Caspase-3表达的影响,以此探讨活血熄风方抗细胞凋亡及对脑缺血的神经保护作用及作用机制.结果:活血熄风方能抑制MCAD模型大鼠大脑皮质Caspase-3蛋白表达.结论:活血熄风方对大鼠局灶性脑缺血具有保护作用,其机理可能与活血熄风方影响抑制细胞凋亡相关蛋白Caspase-3的表达有关.%Objective: To study and explore the neuroprotective effects and the further mechanism of Huoxue Xifeng Decoction on cerebral ischemia of rats.Methods: In order to observe the neuroprotective effect of Huoxue Xifeng Decoction on cerebral ischemia, the rats with incomplete cerebral ischemia were induced with fishing lines in the middle cerebral artery occlusion (MCAO).It was detected the effects of Huoxue Xifeng Decoction on protein expression of cysteiny aspartate specific protease - 3 ( Caspase - 3 ) with immuneohistochemical method.Results: Huoxue Xifeng Decoction can inhibit the expression of Caspase -3 protein.Conclusion: Huoxue Xifeng Decoction had a protective effect on cerebral ischemia and the mechanism is related to inhibiting cell apoptosis and affecting the related expression of Caspase -3 protein interrelated with cell apoptosis.
