粽叶中多糖的提取分离和光谱分析

             

摘要

采用热水浸提法,在90℃条件下水浴回流提取粽叶多糖;以sevag法、透析、醇析、SephadexG-100柱层析对棕叶多糖纯化;以Molish试剂和25%茚三酮乙醇试剂、紫外光谱检测纯化效果;以红外光谱测定粽叶多糖的分子结构.结果表明:用此法提取多糖分离纯化效果较优.红外光谱显示粽叶多糖组分在3600~3200 cm-1、3200~2800 cm-1、1400~1200 cm-1和1200~1000 cm-1 谱段内都出现了典型多糖物质的吸收峰,在890 cm-1的吸收峰表明多糖结构中存在β2糖苷键结构,在820 cm-1处吸收峰有α-D-半乳吡喃糖的特征吸收峰.%By using hot water reflux extraction method, the polysaccharides were extracted from indocalamus in 90℃ water bath; by using sevag law, dialysis, alcohol precipitation, SephadexG ~100 column chromatography to purify polysaccharides extracted from indocalamus; by using Molish reagents, 2.5% of ninhydrin ethanol reagent, and ultraviolet spectrum to verify the effect of purification; by using infrared spectroscopy to determine the molecular structure of polysaccharides from indocalamus. The results showed that using these methods to extraction separation polysaccharides from indocalamus had the best effects; the infrared spectroscopy showed that polysaccharides of indocalamus showed a typical absorption peak in 3 600~3 200 cm~1, 3 200~2 800 cm~1, 1 400~1 200 cm~1, and 1 200~1 000 cm~1. and the absorption peak in the 890 cm~l indicated that there is β2 glycosidic bond structure in polysaccharides structure, which is the characteristic absorption peak of α~D~galactosyl pyranose.
