Cry1Ab蛋白在黑腹果蝇体内的生物富集及对体内3种保护酶活性的影响

     

摘要

采用ELISA技术研究了Cry1Ab蛋白在黑腹果蝇体内的富集情况,并运用分光光度法测定了其体内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和乙酰胆碱酯酶(AchE)活力大小,探讨了Cry1Ab蛋白的富集量和3种保护酶活性的关系.结果表明:在Cry1Ab蛋白培养基上培养1~10 d的果蝇体内存在Cry1Ab蛋白富集,且在第10天富集量达到最高,为对照组的3.7倍.果蝇体内的AchE、SOD和GSH-Px活力均低于对照组,在第5天时GSH-Px受到的抑制作用最强;在第10天时SOD和AchE受到的抑制作用最强;培养10~15 d时,Cry1Ab蛋白富集量逐渐减少,3种酶的活力逐渐增加.Cry1Ab蛋白短时间能在果蝇体内产生富集效应并对果蝇产生一定程度的毒害作用,但果蝇经过一段时间自我保护机制的调节可以逐渐消除Cry1Ab蛋白的毒害作用.%ELISA technique was used to assess the bioaccumulation of CrylAb in the Drosophila melanogaster, and spectrophotometry was used to determine activities of three protective enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and acetylcholinesterase (AchE), and then the relationship between enrichment amount of CrylAb protein and the three protective enzymes was also discussed. The results showed that the enrichment of CrylAb protein existed in the drosophila fed with the CrylAb protein medium for 1-10 days, and the enrichment amount reached the peak at the 10th day which was 3.7 times to the control group. Meanwhile, activities of AchE, SOD and GSH-Px in the drosophila from treatment group were lower than those in the drosophila from control group. In addition, activity of GSH-Px was inhibited the most at the 5th day, and activities of AchE and SOD were inhibited the most at the 10th day. During 10 ~15 days, the enrichment amount of CrylAb protein was gradually reduced, yet activities of the three enzymes reversed. The results also indicated that CrylAb protein produced a toxin effect on the drosophila in a short time, but the drosophila gradually eliminated this toxic effect via the regulation of self-protection mechanism for a period time.
