1,2,3-三氯丙烷对小鼠的亚急性和亚慢性器官损伤的研究

     

摘要

目的 探讨1,2,3-三氯丙烷对Spratue-Dawley大鼠各个系统的亚急性个亚慢性损伤.方法 Spratue-Dawley大鼠200只,1,2,3-三氯丙烷溶于玉米油后经口摄入,大鼠分别经过0.01,0.05,0.2和0.8 mmol·kg-1·d-1,共10 d(亚急性组),和0.01,0.05,0.1,0.4 mmol·kg-1·d-1,共90 d(亚慢性组)的分别处理,每个剂量组20只,雌雄各半.结果 经处理的亚急性和亚慢性组中,体重下降首先出现在高剂量组(P<0.05).当各器官经体重标化后,亚急性和亚慢性的高剂量组,肝脏和肾脏均有增加.在高剂量组别,病理学检查表明,肝功能有轻度受损,肾功能未见毒性反应,心肌细胞的变性坏死呈弥漫性表现,伴有嗜酸性粒细胞增多.结论 本研究表明,心脏和肝脏是1,2,3-三氯丙烷的靶器官,1,2,3-三氯丙烷的暴露与癌变有相关性.%Objective To investigate the subacute and subchronic injury of 1,2,3 TCP on the Sprague Dawley rats' each system. Methods After the 1,2,3 TCP dissolved in corn oil , 200 Dawley rats injected by oral. The subacute groups were fed with the dose of 0. 01,0.05,0. 20 and 0. 80 mmol ·kg-1 · d-1 for 10 days and the suchronic group with 0. 01,0.05 ,0. 10 and 0. 40 mmol · kg-1 · d-1for 90 days, each dose group had 20 rats with female and male in half. Results Among the subacute and subchronic groups, weight lose were observed primarily in the highest doses( P <0. 05 ). When major organ weights were normalized by body weight, liver and kidney values were generally increased in the two highest dose groups. The pathological examination showed, in the high doses, the liver function were injuried slightly, while no toxic reaction happened in the kidney function. Myocardial necrosis and degeneration occurred in a diffuse pattern with marked eosinophilia of affected cells. Conclusion This study demonstrates that heart and liver are target organs for 1,2,3 Trichloropropane and suggeststhat 1,2,3 Trichloropropane exposure may be related to canceration.
