补阳还五汤对高脂血症大鼠血流变及血清一氧化氮的影响

     

摘要

目的 研究补阳还五汤对高脂血症大鼠血液流变学及血清一氧化氮(NO)的影响.方法 将健康SD大鼠60只随机分为6组,即空白对照组、模型组和补阳还五汤高、中、低剂量组以及血脂康组,除空白对照组大鼠常规饲料喂养外,其余大鼠喂养高脂饲料,同时补阳还五汤高、中、低剂量组分别灌服补阳还五汤28、21、14 g生药/kg,血脂康组灌服血脂康0.12 g/kg,空白对照组、模型组灌服等容量0.9%氯化钠注射液.连续喂养8周.末次给药后,禁食12 h,所有动物断头取血,检测总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、全血黏度、红细胞比积(HCT)、红细胞沉降率(ESR)、纤维蛋白原(FIB)及NO.结果 补阳还五汤高、中、低剂量组TC、TG、LDL-C含量与模型组比较明显降低(P<0.01),HDL-C明显升高(P<0.01,P<0.05);补阳还五汤高、中、低剂量组在升高HDL-C方面优于血脂康组(P<0.01);与模型组比较,补阳还五汤高、中、低剂量组全血黏度(高切、低切)均明显降低(P<0.01);HCT明显降低(P<0.01),ESR明显减慢(P<0.01),FIB明显降低(P<0.01);补阳还五汤高、中剂量组在降低纤维蛋白原方面优于血脂康组(P<0.01);补阳还五汤高、中、低剂量组血清NO含量与模型组比较差异有统计学意义(P<0.01),补阳还五汤中、高剂量组NO含量与血脂康组比较差异有统计学意义(P<0.01).结论 补阳还五汤可以改善高脂血症大鼠血液浓、黏、凝、聚状态,具有提高血管内皮功能,减轻动脉粥样硬化的作用.%Objective To observe the effect of Buyanghuanwu decoction Buyanghuan decoction on hemorrheology and serum Nitric oxide in rat with hyperlipoidemia. Methods 60 SD rats were randomly divided into six groups, control group, model group, high-, middle-, low-dose Buyanghuan decoction groups and Xuezhikang group. Besides rat in control group were fed by routine feed, rat in other groups were fed by high fat feed (2% cholesterol, 0.2% propylthiouracil, 0.2% sodium cholate, 10% lard, 87.6% routine feed). Simultaneously rat in high-, middle-, low-dose Buyanghuan decoction groups were fed by 28, 21,14 g/kg Buyanghuan decoction. Rat in Xuezhikang group were fed by Xuezhikang, 0.12g/kg. Eight weeks feed was carried on. All rats were decapitated for collection of blood of rat. TC, TG, HDL-C, LDL-C, blood viscosity, erythrocrit, blood sedimentation rate (BSR), fibrinogen and NO were measured. Results Compared with model group, TC, TG and LDL-C in high-, middle-, low-dose Buyanghuan decoction groups were obviously decreased (P<0.01), HDL-C was increased (P<0.05). Improvement of HDL-C in high-, middle-, low-dose Buyanghuan decoction groups was superior to that in Xuezhikang group (P<0.01). Blood viscosity under high shear rate and low share rate in high-, middle-, low-dose Buyanghuan decoction groups was decreased comparison with model group (P<0.01). Erythrocrit in Buyanghuan decoction groups were obviously decreased (P<0.01), BSR was significantly slowed (P<0.01), fibrinogen was decreased (P<0.01). The effect of decreasing fibrinogen in Buyanghuan decoction groups was superior to that in Xuezhikang group (P<0.01). There was obvious difference between high-, middle-dose Buyanghuan decoction groups and Xuezhikang group in content of NO (P<0.01). Conclusion Buyanghuanwu decoction can improve thickness, sticky, coagulation and gather of blood in rat with hyperlipoidemia, enhance vascular endothelial function and relief atherosclerosis effect.
