氧化苦参碱抗B16黑色素瘤的实验研究

     

摘要

Objective To investigate the effects of oxymatrine on malignant melanoma B16 cells. Methods B16 cells were cultured with oxymatrine (0.5、1.0、2.0、4.0、8.0 mg/mL) for 24 h, and the rates of growth inhibition were tested by MTT assay. C57BL/6J mice with melanoma tumor were randomized 4 groups: negative control and oxymatrine high, middle, low dose group. Mice were fed with 30、60、120 mg/kg doses of oxymatrine, to monitor the tumor growth, the index of spleen and thymus. Results MTT method indicated that the proliferation of B16 cells was inhibited by oxymatrine at different concentrations. The weight of tumor was decreased, the index of spleen and thymus were increased by oxymatrine. Conclusion Oxymatrine has obvious antitumor effect by inhibiting cell proliferation, regulating abnormal immune state of tumor - loaded mice.%目的 观察氧化苦参碱对B<,16>黑色素瘤的作用.方法 B<,16>细胞分别加入0.5、1.0、2.0、4.0、8.0 mg/,mL氧化苦参碱,孵育24 h,噻唑蓝(MTT)比色法观察不同浓度氧化苦参碱对离体培养的B<,16>黑色素瘤细胞增殖的影响;将B<,16>荷瘤小鼠随机分为对照组及氧化苦参碱高、中、低剂量组,接种后第2 d开始给药,氧化苦参碱高、中、低剂量组分别灌胃给予120、60、30 mg/kg的氧化苦参碱,对照组给予等容积0.9%氯化钠注射液,连续10 d后,处死动物,测定肿瘤生长抑制率、脾脏指数和胸腺指数.结果 MTT比色法显示氧化苦参碱对B<,16>黑色素瘤细胞具有明显的增殖抑制作用;氧化苦参碱能明显减轻小鼠瘤质量,升高脾脏指数、胸腺指数.结论 氧化苦参碱可通过抑制肿瘤细胞增殖、调节荷瘤机体异常的免疫状态而发挥抗B<,16>黑色素瘤作用.
